中枢神经系统感染易感性与MMP-2和VEGF基因多态性的相关性 被引量:1

Correlation between susceptibility to central nervous system infection and gene polymorphisms of MMP-2 and VEGF

摘要 目的分析基质金属蛋白酶-2(MMP-2)和血管内皮生长因子(VEGF)基因多态性与中枢神经系统感染易感性的相关性。方法选取2017年1月-2019年12月北部战区总医院收治的中枢神经系统感染患者96例为病例组,30例未发生感染颅脑损伤患者为对照组。比较两组患者脑脊液和血清中MMP-2、VEGF水平差异,聚合酶链式反应(PCR)检测MMP-2-735位点、MMP-2-1306位点、VEGF-460位点、VEGF+936位点的基因型。结果病例组患者MMP-2脑脊液和血清水平分别为(243.82±47.24、1056.80±229.45)μg/L,VEGF脑脊液和血清水平分别为(66.23±15.24、85.04±20.11)ng/L均高于对照组(P<0.05);病例组MMP-2-735 C/T位点和-1306 C/T位点的CC基因型频率高于对照组,CT基因型频率低于对照组(P<0.05),且C等位基因频率高于对照组,T等位基因频率低于对照组(P<0.05);病例组VEGF-460 C/T位点、+936 C/T位点的CC基因型频率低于对照组,TT基因型频率高于对照组(P<0.05),且C等位基因频率低于对照组,T等位基因频率高于对照组(P<0.05)。结论中枢神经系统感染患者脑脊液和血清中MMP-2、VEGF水平均升高,MMP-2-735 C/T位点、MMP-2-1306 C/T位点、VEGF-460 C/T位点、VEGF+936 C/T位点的基因多态性与中枢神经系统感染疾病易感性有关。 OBJECTIVE To analyze the correlation between gene polymorphisms of matrix metalloproteinase-2(MMP-2)and vascular endothelial growth factor(VEGF)and the susceptibility to central nervous system infection.METHODS Totally 96 patients with central nervous system infection who were treated in the North Theater General Hospital from Jan.2017 to Dec.2019 were assigned as the case group,and 30 craniocerebral injury patients without infection were set as the control group.The levels of MMP-2 and VEGF in cerebrospinal fluid and serum were compared between the two groups of patients,and the genotypes at MMP-2-735 locus,MMP-2-1306 locus,VEGF-460 locus and VEGF+936 locus were detected by means of polymerase chain reaction.RESULTS The cerebrospinal fluid MMP-2 level of the case group was(243.82±47.24)μg/L,the serum MMP-2 level(1056.80±229.45)μg/L;the cerebrospinal fluid VEGF level of the case group was(66.23±15.24)ng/L,the serum VEGF level(85.04±20.11)ng/L;the levels of above indexes of the case group were significantly higher than those of the control group(P<0.05).The frequencies of CC genotype at MMP-2-735 C/T locus and-1306 C/T locus were significantly higher in the case group than in the control group,while the frequencies of CT genotype were significantly lower in the case group than in the control group(P<0.05),the frequency of C allele of the case group was significantly higher than that of the control group,and the frequency of T allele of the case group was lower than that of the control group(P<0.05).The frequencies of CC genotype at VEGF-460 C/T locus and+936 C/T locus were significantly lower in the case group than in the control group,while the frequencies of TT genotype were significantly higher in the case group than in the control group(P<0.05),the frequency of C allele of the case group was significantly lower than that of the control group,and the frequency of T allele of the case group was significantly higher than that of the control group(P<0.05).CONCLUSION The levels of MMP-2 and VEGF in both cerebrospinal fluid and serum of the patients with central nervous system infection are elevated,and the gene polymorphisms at MMP-2-735 C/T locus,MMP-2-1306 C/T locus,VEGF-460 C/T locus and VEGF+936 C/T locus are associated with the susceptibility to the central nervous system infection.
