摘要
本研究对青海省西宁市野生动物园雪豹发生的疑似水貂肠炎病毒(MEV)腹泻粪便样本利用分子生物学方法进行了鉴定。结果显示:共成功鉴定到两株MEV(QH1/2),并对MEV的NS1基因进行了克隆测序分析,两株病毒株间核苷酸序列和氨基酸序列同源性分别为99.65%和99.55%;MEV-QH1/2病毒株与其他株间NS1氨基酸比对有19个氨基酸位点存在变异,但不存在特异性变异位点,两株病毒株间仅有3个变异位点,分别位于350、574和595位。进化树显示:MEV-QH1和MEV-QH2聚在一个大的分支上,但并未聚在一个微小分支上,这表明两株病毒株间存在一定的遗传变异。本研究对我国青藏高原雪豹源MEV进行了分子鉴定与遗传变异分析,丰富了MEV的遗传学研究资料。
Molecular biological methods are used in this study to identify fecal samples of suspected mink enteritis virus(MEV)diarrhea of snow leopard in Xining Wildlife Park of Qinghai province.The results show that two strains of MEV(QH1/2)are identified and NS1 gene of MEV is cloned and sequenced.The homology of nucleotide sequence and amino acid sequence is 99.65%and 99.55%respectively;compared uith other strains,there are 19 amino acid sites of NS1 in MEV-QH1/2 strains uithout specific mutation sites.Only 3 mutation sites are found between two strains,namely position 350,574 and 595.Phylogenetic tree shows that MEV-QH1 and MEV-QH2 are clustered on a large branch,but not on a small branch,indicating that there exists certain genetic variation between the two strains.Molecular identification and genetic variation analysis of MEV from snow leopard in Qinghai-Tibet Plateau are carried out in this paper,which enriches the genetic research materials of MEV.
