甘蔗新基因ShUGT2的生物信息学分析及亚细胞定位 被引量:2

摘要 UDP-糖基转移酶在植物体内负责糖基化修饰,对植物的生长发育起着关键作用。以甘蔗品种ROC22为供试材料,提取成熟叶片总RNA并反转录合成cDNA,经PCR扩增获得ShUGT2基因cDNA全长序列,对其进行生物信息学、亚细胞定位和组织特异性表达分析。结果表明:ShUGT2含有1782 bp的完整开放阅读框,编码593个氨基酸,其编码蛋白与高粱UDP-糖基转移酶同源性达到92.45%。ShUGT2编码蛋白不稳定系数为48.68,疏水性指数-0.338,是一种亲水性蛋白;在20~42个氨基酸的位置,有一个跨膜区;在309~514个氨基酸,存在UDP糖基转移酶的家族特异保守结构域,属于GT8家族成员,其功能是通过合成木聚糖,调控细胞内半纤维素成分,维持植物的生长发育。亚细胞定位分析表明,ShUGT2编码蛋白定位于线粒体;在甘蔗的根、茎、叶组织中,ShUGT2基因均有表达,与甘蔗的生长发育密切相关。本研究结果为进一步分析ShUGT2基因功能提供了科学依据。 UDP-glycosyltransferase plays a key role in development and growth of plants through glycosylation modification.The total RNA was extracted from mature leaves of sugarcane cultivar ROC22,and the first-strand cDNA was synthesized.Then,the full-length cDNA sequence of ShUGT2 gene was obtained by PCR.Furthermore,the bioinformatics,encoding protein and expression levels of ShUGT2 were analyzed.The analysis showed that ShUGT2 has a 1782 bp complete open-reading frame and encodes 593 amino acids.Its encoding protein has 92.45%homology with that of sorghum UDP-glycosyltransferase.The ShUGT2 encoding protein has an instability coefficient of 48.68 with a hydrophobicity index being-0.338.This indicates that the ShUGT2 encoding protein is a hydrophilic protein with a transmembrane domain at 20~42 amino acids position.There is a conservative domain of UDP-glycosyltransferase family at 309~514 amino acids position,a member of the GT8 family whose function is to regulate intracellular hemicellulose components and maintain plant growth and development by synthesizing xylan.The subcellular localization analysis showed that the ShUGT2 encoding protein is located in mitochondria.The ShUGT2 gene is expressed in roots,stems and leaves of sugarcane,which is closely related to the growth and development of sugarcane.This study provides a scientific basis for further analysis of ShUGT2 gene functions.
