养血柔肝针法加减对围绝经期抑郁症小鼠雌激素水平的影响

Effects of modified YangXueRouGan acupuncture on estrogen level in perimenopausal depression mice

目的探讨养血柔肝针法加减对围绝经期抑郁症小鼠雌激素(E2)水平、阴道脱落细胞上皮角化细胞对E2的阳性反应率及子宫指数的影响。方法 2020年8—10月,通过将雌性小鼠去除卵巢后进行冰水游泳刺激制备围绝经期抑郁症的动物模型,选用SPF级5~6周未孕昆明种小鼠30只,实验分成3组,每组10只,包括针刺组、去势对照组、正常对照组。3组小鼠每天均进行冰水游泳刺激。针刺组进行养血柔肝针法加减处理,治疗时间为4周,随后计算小鼠的阴道脱落细胞角化细胞对E2的阳性反应率、子宫指数,并检测血清E2水平。结果与正常对照组比较,去势对照组的阴道脱落细胞上皮角化细胞对E2的阳性反应率(0 vs 100.00%,P<0.01)、子宫指数(0.56±0.11 vs 1.55±0.28,P<0.01)及血清E2水平[(9.35±0.13)pg/mL vs (20.00±0.22)pg/mL,P<0.01]明显降低。与去势对照组比较,针刺组的阴道脱落细胞上皮角化细胞对E2的阳性反应率(77.78%vs 0,P<0.01)、子宫指数(1.45±0.15 vs 0.56±0.11,P<0.01)及血清E2水平[(17.00±0.18)pg/mL vs(9.35±0.13)pg/mL,P<0.01]明显升高。结论养血柔肝针法加减可以提高围绝经期抑郁症小鼠的E2水平阴道脱落细胞上皮角化细胞对E2的阳性反应率和子宫指数。

Objective To explore the effects of the modified YangXueRouGan acupuncture on the estrogen(E2) level,the positive reaction rate of exfoliated vaginal epithelial keratinocytes to E2 and the uterine index in perimenopausal depression mice.Methods The animal model of perimenopausal depression was established by ovariectomy followed by ice-water swimming stimulation from August to October 2020. 30 SPF Kunming unpregnancy mice aged 5-6 weeks were separated into three groups:acupuncture group,ovariectomy group,and control group,10 animals in each group. Ice-water swimming stimulation was carried out in three groups every day. The modified YangXueRouGan acupuncture was performed in the acupuncture group. The treatment duration was four weeks. The positive reaction rate of exfoliated vaginal epithelial keratinocytes to E2 and the uterine index were calculated,and the serum E2 level was detected. Results Compared with the normal control group,the positive reaction rate of exfoliated vaginal epithelial keratinocytes to E2(0 vs 100.00,P<0.01),uterine index(0.56±0.11 vs 1.55±0.28,P<0.01),and serum E2 level(9.35±0.13 vs 20.00±0.22,P<0.01) in the ovariectomy group were significantly reduced. Compared with the ovariectomy group,the positive reaction rate of exfoliated vaginal epithelial keratinocytes(77.78 vs 0,P<0.01),uterine index(1.45 ±0.15 vs0.56±0.11,P<0.01),and serum E2 level(17.00±0.18 vs 9.35±0.13,P<0.01) in the acupuncture group were significantly increased.Conclusion The modified YangXueRouGan acupuncture can improve the positive reaction rate of exfoliated vaginal epithelial keratinocytes to E2,E2 level and uterine index in perimenopausal depression mice.
