华蟹甲的简化基因组测序及SSR引物开发 被引量:1

摘要 中国特有的菊科植物华蟹甲是潜在的抗肿瘤、抗氧化、抗菌等特效药物资源,同时也是具有潜在入侵性的一种大型草本植物.通过简化基因组测序的方法,对华蟹甲的SSR信息进行全面的分析,并据此开发其SSR分子标记.结果表明:华蟹甲简化基因组SSR位点数量和类型丰富,共得到双端含有100 bp六种类型的SSR(一至六核苷酸)的SSR位点46674个,其中包含均有引物片段的SSR数目为43235,片段引物设计率为92%.单核苷酸重复、二核苷酸重复和三核苷酸重复类型的数量较多,占主导地位.在321种重复单元中,最常见的是单核苷酸重复(A/T)10.利用已合成的200对引物对16个华蟹甲样品进行荧光SSR扩增,共筛选出20对特异性和多态性较高的引物.本研究通过华蟹甲简化基因组测序进行SSR引物开发,为华蟹甲遗传结构、图谱构建、标记辅助育种以及物种管理等方面提供依据并奠定了基础. Sinacalia tangutica(Asteraceae),the tall herb endemic to China,is very important for its therapeutic potential as anti-tumor,anti-oxidation and anti-bacterial activities,as well as the potential invasiveness.Here,Reduced-Representation Genome Sequencing(RRGS)was applied to Sinacalia tangutica(S.tangutica)in order to rapidly and systematically identify SSR markers.Results:SSR loci with 1-6 bp repeat motifs were identified from the sequencing data,and a total of 46674 SSR loci were identified.Among the SSR loci,mononucleotide SSRs were the most abundant microsatellite types(21529,46.13%),followed by dinucleotide microsatellites(18426,39.48%).Of the 321 possible motifs,AT/TA motifs were most frequently represented.Finally,20 polymorphic SSR markers were isolated and characterized from 200 primers pairs designed.This study is the key step toward the further studies on genetic diversity and structure,which will play important roles in species conservation and management in S.tangutica.
