沙门菌噬菌体混合制剂研制及体外杀菌效果评价 被引量:1

摘要 为开发一种可用于体内或体外沙门菌防控的噬菌体混合制剂,使用丝裂霉素C从不同血清型沙门菌中诱导并纯化获得沙门菌噬菌体,初步分析其裂解谱和理化条件适应性。为拓展噬菌体制剂的裂解谱并综合发挥温和噬菌体pP14及烈性噬菌体pSQC73的优势,制成噬菌体混合制剂并评价其体外杀菌抑菌效果。结果表明:沙门菌噬菌体pP14可耐受50℃高温,在pH值4~12范围内活性均较稳定,和pSQC73混合制成的制剂在3 h内可有效减少鼠伤寒沙门菌和鸡白痢沙门菌的数量,在1 h内可使细菌数量降低90%以上,针对低浓度的细菌时,稳定杀菌抑菌时间可维持3~6 h。该制剂对体外畜禽养殖环境中常见沙门菌血清型具有较好的杀灭和抑制效果,有望作为环境杀菌剂甚至体内杀菌药物的开发潜力。 To develop a Salmonella bacteriophage cocktail preparation for utilizing as in vitro or in vivo disinfection reagent in controlling Salmonella, Salmonella phages from different Salmonella serotype strains were successfully induced and purified using Mitomycin C, and their biological characteristics in different temperature and pH were analyzed. To combine the lysis spectrum and advantages of temperate phage and virulent phage, the pP14 and pSQC73 bacteriophage were mixed to be a cocktail preparation and its bactericidal effect in vitro was evaluated. The results showed that the pP14 bacteriophage can keep its viability under the condition of 50 ℃ and pH 4-12. The cocktail mixture of pP14 and pSQC73 bacteriophage reduced the number of Salmonella Typhimurium and Salmonella Pullorum by more than 90% within 1 h. This high-efficiency bactericidal effect can be maintained for 3-6 hours when treated against low concentration of bacteria. This study indicated that the developed cocktail mixture has a better inhibiting effect on common Salmonella serotypes in livestock and poultry breeding environments.
