当归补血汤对糖尿病足大鼠模型血管生成和炎症反应的影响 被引量:13

Effect of Danggui Buxue Decoction on Angiogenesis and Inflammatory Reaction in Rat Models with Diabetic Foot

摘要 目的研究当归补血汤治疗糖尿病足大鼠模型的效果,以及对血管生成和炎症反应的影响。方法选取成年健康SD大鼠,给予高脂饲料饲养1个月,应用链脲佐菌素建立糖尿病大鼠模型,并在足部造矩形切口建立糖尿病足大鼠模型。选择正常大鼠10只为对照组,糖尿病足模型大鼠40只,随机分为模型组、低、中和高剂量组,每组10只。低、中、高剂量组分别给予生药浓度为2、4、8 g/kg当归补血汤灌胃,对照组和模型组生理盐水灌胃,每天1次,持续1周。观察5组足部创面愈合情况,比较血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、IL-1β和血管内皮生长因子(VEGF)含量,同时比较新生肉芽组织TNF-α、IL-6、IL-1β和VEGF mRNA表达水平。结果模型组创面面积大于对照组,血清TNF-α、IL-6、IL-1β含量及新生肉芽组织TNF-α、IL-6、IL-1βmRNA表达水平均高于对照组,血清VEGF含量及新生肉芽组织VEGF mRNA表达水平低于对照组(P<0.01)。低、中、高剂量组创面面积均小于模型组,血清TNF-α、IL-6、IL-1β含量及新生肉芽组织TNF-α、IL-6和IL-1βmRNA表达水平均低于模型组,血清VEGF含量及新生肉芽组织VEGF mRNA表达水平均高于模型组,并具有剂量效应(P<0.05,P<0.01)。结论当归补血汤能降低糖尿病足大鼠模型炎症反应,促进新生血管生成,加快创面愈合。 Objective To study effectiveness of Danggui Buxue Decoction in treatment of rat models with diabetic foot and its effects on angiogenesis and inflammatory reaction.Methods Healthy adult SD rats were selected and fed with high fat diet for one month.Streptozotocin was used to establish rat models with diabetic,and rectangular incisions were made in the feet to establish rat models with diabetic foot.Another 10 healthy rats were selected as control group,and 40 rat models with diabetic foot were randomly divided into model group(n=10),low dose group(n=10),moderate dose group(n=10)and high dose group(n=10).Low,moderate and high dose groups were given Danggui Buxue Decoction with concentrations of 2,4 and 8 g/kg respectively,while control and model groups were treated with physiological saline by gastric lavage once a day for one week.Foot conditions of wound healing were observed,and contents of serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),IL-1βand vascular endothelial growth factor(VEGF)were compared among 5 groups.In the meantime,mRNA expression levels of TNF-α,IL-6,IL-1βand VEGF in new granulation tissues were also compared among 5 groups.Results In model group,wound area was significantly larger,and serum contents of TNF-α,IL-6 and IL-1βand mRNA expression levels of TNF-α,IL-6 and IL-1βin new granulation tissues were significantly higher,while serum content of VEGF and mRNA expression level of VEGF in new granulation tissues were significantly lower than those in control group(P<0.01).In low,moderate and high dose groups,wound areas were significantly smaller,and serum contents of TNF-α,IL-6 and IL-1βand mRNA expression levels of TNF-α,IL-6 and IL-1βin new granulation tissues were significantly lower,while serum content of VEGF and the mRNA expression level of VEGF in new granulation tissues were significantly higher than those in model group(P<0.05,P<0.01),and the differences showed a dose-effect relationship(P<0.05,P<0.01).Conclusion Danggui Buxue Decoction may reduce inflammatory reaction,promote neovascularization and accelerate wound healing in rats with diabetic foot.
