摘要
目的基于线粒体基因组数据探讨库蚊亚属(Culicidae,Culex)系统发育关系。方法整合13个蛋白编码基因序列,分别构建库蚊亚属19个物种的系统发育树,利用MEGA7.0软件估算物种间的遗传距离。结果系统发育分析表明,仅Pipiens亚组/复合组的单系性得到支持,Sitiens组、Coronator组、及Pipiens组内其他亚组/复合组的单系性均不成立。遗传距离说明不同物种间亲缘关系的远近,其中Tarsalis亚组是分化最明显的一个类群,该亚组的物种分别与Coronator组、Apicinus亚组的物种聚成不同分支,且该亚组的Culex lygrus与其他物种间的遗传距离均>0.04,在系统数中单独成为一支。结论库蚊亚属种类繁多,但多数亚组/复合组不具单系性,个别亚组已分化为单独类群,因此可在形态特征和生物学特性的基础上,结合分子生物学方面的证据,确保物种划分的准确性。
Objective To examine phylogenetic relationships among species of subgenus Culex based on its mitochondrial genomes.Methods Phylogenetic trees were constructed and genetic distances were evaluated for 19 species of the subgenus Culex based on the combined data on sequences of 13 genes that code for proteins.Results Phylogenetic analysis revealed support for the monophyly of the Pipiens subgroup/complex.Groups including Sitiens,Coronator,and Pipiens were not supported.Genetic distances further indicated differences among species.Species of the Tarsalis subgroup exhibited obvious divergence.Species of this subgroup were clustered into the Coronator group and Apicinus subgroup.In addition,the genetic distance between Culex lygrus and all other species exceeded 0.04,and C.lygrus form a separated clade in the phylogenetic tree.Conclusion Classification of the subgenus Culex is needs to be confirmed.Further evidence should be compiled,like morphological and biological characteristics and molecular biological information.
