头针抑制脑出血后神经细胞凋亡机制初探 被引量:9

摘要 目的:观察“百会”透“曲鬓”针刺法对脑出血大鼠出血半暗带神经细胞凋亡的影响及Shh信号通路在此过程发挥的作用。方法:健康雄性SD大鼠随机分为假手术组、模型组、模型+针刺组(针刺组)、模型+激动剂组(激动剂组)、模型+非经非穴组(非经非穴组),每组各15只。将50μL自体血注入右脑尾状核复制脑出血模型(假手术组以同剂量生理盐水代替),随后每日1次“百会”透“曲鬓”针刺治疗。连续干预5 d后,Bederson神经功能评分观察大鼠神经功能恢复,HE染色观察出血半暗带病理损伤,TUNEL检测凋亡神经细胞,Western blot检测Shh通路Smo、Gli1蛋白表达。结果:针刺可明显促进脑出血大鼠神经功能恢复,改善出血半暗带病理损伤,下调该区凋亡细胞率,并促进Smo、Gli1蛋白表达。结论:“百会”透“曲鬓”针刺法可明显促进脑出血大鼠神经功能恢复,这可能与其激活Shh通路,继而抑制脑出血半暗带区神经细胞凋亡有关。 Objective:To observe the effect of needling from Baihui(DU20)to Qubin(GB7)on neural cells apoptosis at hemorrhagic penumbra after intracerebral hemorrhage(ICH)and the effect of Shh pathway on this progress.Methods:Male SD rats were randomly divided into the sham group,the model group,the acupuncture group,the agonist group and the sham acupuncture group,with 15 rats in each group.ICH model was established by injecting 50μl autogenous blood into right caudate nucleus,and the rats in the sham group were injected with 50μl saline instead.Then rats were treated with needling from DU20 to GB7 each day.After 5 consecutive days of intervention,neurological defect was scored with Bederson's method,HE assay was used to observe pathological injuries at hemorrhagic penumbra,TUNEL assay was used to detect apoptosis cells,and Western blot was used to measure protein expressions of Smo and Gli1.Results:Acupuncture could remarkably improve neural functions and pathological injuries at hemorrhagic penumbra,decrease the apoptosis rate of the cells at hemorrhagic penumbra,and also activate protein expressions of Smo and Gli1 after ICH.Conclusion:Needling from Du20 to GB7 can obviously improve neural functions after ICH,which is related to activating Shh pathway and in further inhibiting neural cells apoptosis.
