摘要
目的:探索活血散结中药复方含药血浆对碱性成纤维细胞生长因子(bFGF)干预下兔视网膜色素上皮(RPE)细胞增殖的影响。方法:选用对数生长期兔RPE细胞,分成空白对照组、正常血浆组、bFGF组、bFGF+BGJ398组、bFGF+含药血浆组5组,CCK法测定各组兔RPE细胞的细胞活力,流式法检测各组兔RPE细胞的凋亡率。结果:与bFGF组比较,bFGF+BGJ398组、bFGF+含药血浆组细胞活力显著抑制(P<0.01);细胞凋亡率显著增加(P<0.01)。结论:活血散结中药复方含药血浆能有效抑制bFGF干预下兔RPE细胞增殖,促进兔RPE细胞的凋亡。
Objective:To explore the inhibition effect of invigorating blood and dissipating masses Chinese medicine compound drug-cotaining plasma on rabbit retinal pigment epithelium(RPE)cell proliferation under intervention of basic fibroblast growth factor(bFGF).Methods:Select the rabbit RPE cells in log phase.The experiment objects were divided into the blank control group,the normal plasma group,the bFGF group,the bFGF+BGJ398 group,the bFGF+drug-containing plasma group.CCK method was used to detect the absorbance value(OD)of rabbit RPE cells from different groups.The flow method was adopted to measure the apoptosis rate of rabbit RPE cells in different groups.Results:Compared with bFGF group,the cell activity of bFGF+BGJ398 group and bFGF+drug-containing plasma group were significantly inhabited(P<0.01);The apoptosis rate was increased evidently(P<0.01).Conclusion:Invigorating blood and dissipating masses Chinese medicine compound drug-cotaining plasma can inhibit the proliferation and promote the apoptosis of rabbit RPE cells under bFGF effect.
