摘要
为进行鸡腿菇(Coprinus comatus)的种质资源鉴定和及其分子育种提供理论依据,以22株鸡腿菇菌株为试材,采用ISSR和SRAP分子标记技术,对样本进行遗传多样性分析。采用筛选出的24条ISSR引物和24对SRAP引物以22株鸡腿菇的基因组DNA为模板进行PCR扩增,共获得清晰的条带368条,其中多态性条带331条占比89.9%。对获取的条带进行UPGMA聚类分析表明,在遗传相似系数为0.8水平上,可将22株供试鸡腿菇菌株分为4大类,明确了菌株之间的亲缘关系;聚类分析与主坐标分析基本一致;22株供试鸡腿菇的遗传距离在0.0300~0.8503;上述结果表明22株供试鸡腿菇菌株间具有较大的遗传差异和多样性。
In order to provides a theoretical basis for the identification of Coprinus comatus germplasm resources and molecular breeding,22 C.comatus strains were used as test materials,using ISSR and SRAP molecular marker technology,the genetic diversity of 22 C.comatus samples collected across the country was analyzed.The 24 ISSR primers and 24 pairs of SRAP primers selected were used for PCR amplification using the genomic DNA of 22 C.comatus as a template.A total of 368 clear bands were obtained,of which 331 polymorphic bands accounted for 89.9%.UPGMA clustering of the obtained bands showed that 22 tested C.comatus strains can be divided into 4 categories at a genetic similarity coefficient of 0.8,the cluster analysis is basically the same as the principal coordinate analysis.The genetic distance of C.comatus is between 0.0300-0.8503,and the above results indicate that the 22 tested C.comatus strains have greater genetic differences and diversity.
