摘要
目的研究肿瘤相关基因P53、PTEN、c-Myc在乳腺癌和良性乳腺疾病患者外周血白细胞中的表达情况,探讨其与临床病理资料间的相互关系及其潜在的临床应用价值。方法采用实时荧光定量PCR法检测304例研究对象外周血白细胞中P53、PTEN、c-Myc mRNA相对表达水平,其中乳腺癌组105例,良性乳腺疾病组107例,对照组92例。采用2-ΔΔCT相对定量法对各基因表达情况进行相对定量,使用Mann-whitney U检验比较各组间基因表达差异。结果乳腺癌组和良性乳腺疾病组中PTEN和P53 mRNA相对表达水平均低于对照组,差异有统计学意义(P<0.05);良性乳腺疾病组P53 mRNA相对表达水平明显低于乳腺癌组,差异有统计学意义(P<0.05);各组c-Myc mRNA相对表达水平比较,差异无统计学意义(P>0.05)。乳腺癌组淋巴结转移阴性患者外周血白细胞中PTEN mRNA相对表达水平明显低于淋巴结转移阳性患者,差异有统计学意义(P<0.05)。结论PTEN mRNA相对表达水平下调能预警更为早期的乳腺癌发生,对肿瘤的早期和无创诊断极具潜在的临床应用与推广价值。
Objective To detect the expression of P53,PTEN,c-Myc mRNA in peripheral blood leukocytes from patients with breast cancer and benign breast diseases,and investigate its correlation with the clinicopathologic characteristics,further to explore the potential clinical application significance.Methods Real-time quantitative PCR(RT-qPCR)was employed in this study to detect the expression of P53,PTEN,c-Myc mRNA in peripheral blood leukocytes from 304 cases,including 105 patients with breast cancer group,107 cases of benign breast diseases group and 92 healthy women as control group.2-ΔΔCT was used to relative quantitative the level of gene expression and Mann-whitney U test was used to compare the expression of gene among groups.Results The mRNA relative expression levels of PTEN and P53 in breast cancer group and benign breast disease group were significantly lower than those in control group,with statistical significance(P<0.05).The relative expression level of P53 mRNA in benign breast disease group was significantly lower than that in breast cancer group,and the difference was statistically significant(P<0.05).There was no statistical significance in the relative expression level of c-Myc mRNA among all groups(P>0.05).The relative expression level of PTEN mRNA in peripheral white blood cells of the breast cancer group with lymph node metastasis negative was significantly lower than that of the breast cancer group with lymph node metastasis positive(P<0.05).Conclusion The downregulation of PTEN mRNA relative expression level can predict the early occurrence of breast cancer,and has potential clinical application and promotion value for the early and non-invasive diagnosis of tumor.
