不同发育阶段云芝子实体水提物保肝作用 被引量:2

Hepatoprotective Effect of C.versicolor Fruiting Body Water Extracts at Different Developmental Stages

摘要 对不同发育阶段云芝(Coriolus versicolor)子实体(黑色、黄色、青色云芝)的多糖含量进行测定;通过连续灌胃56度白酒建立小鼠肝损伤模型,研究云芝子实体水提物(低、中、高剂量分别为1.35、2.70、4.05 g·kg^(-1)·d^(-1))对小鼠醉酒入睡时间,肝脏指数,血清中尿素、谷草转氨酶、谷丙转氨酶、白蛋白、总蛋白含量的影响;并采用HE染色观察肝组织的病理变化。结果表明:黄色、青色云芝多糖含量分别显著、极显著高于黑色云芝。与模型组比较,水提物各组小鼠醉酒入睡时间均显著延长、肝脏指数均极显著下降、血清总蛋白含量均显著增加;除了黑色云芝中、高剂量组外,其他水提物组小鼠血清尿素含量均显著增加;黑色云芝低剂量组、黄色云芝高剂量组、青色云芝各组小鼠血清谷草转氨酶含量均显著增加;黑色云芝低剂量组、黄色云芝中剂量组、青色云芝各组小鼠血清谷丙转氨酶含量均显著增加,其中青色云芝中剂量组增加最为显著;除黑色云芝高剂量组外,其他水提物组小鼠血清白蛋白含量均增加。肝组织切片显示水提物对小鼠肝损伤有一定的修复作用。 Mice were randomly divided into 12 groups with 5 male and 5 female mice in each group as follows:control group,model group,positive group,black C.versicolor stage water extract groups(low,medium and high doses),yellow C.versicolor stage water extract groups(low,medium and high doses),and cyan C.versicolor stage water extract groups(low,medium and high doses).The liver injury model was established by gastric administration of 56 degree liquor at a dose of 12 mL·kg^(-1)·d^(-1)every other day for 14 d.For mice in different water extract treatment groups,they were administered low(1.35 g·kg^(-1)·d^(-1)),medium(2.70 g·kg^(-1)·d^(-1)),and high(4.05 g·kg^(-1)·d^(-1))doses of each extract daily by gavage for 14 d since day 8 of liquor treatment.Mice in the control group and positive group were administered saline and bifendate by gavage,respectively.After different treatments,mice in each group were measured for time to fall asleep,liver index,and serum urea,aspertate aminotransferase(AST),alanine aminotransferase(ALT),albumin(ALB)and total protein levels;pathological changes of liver tissue were observed by hematoxylin-eosin(HE)staining.The results showed that both yellow and cyan C.versicolor contained more polysaccharides than black C.versicolor.Compared with the model group,mice in all the water extract groups took longer time to fall asleep with a decrease in liver index.Except medium-dose and high-dose black C.versicolor water extract groups,mice in all the other water extract groups had an increase in serum urea level compared with the model group.Serum AST was significantly increased in low-dose black C.versicolor,high-dose yellow C.versicolor and all cyan C.versicolor water extract groups compared with the model group.Serum ALT was significantly increased in low-dose black C.versicolor,medium-dose yellow C.versicolor and all cyan C.versicolor water extract groups compared with the model group.Serum ALB was increased in all water extract groups except the high-dose black C.versicolor water extract group.Liver tissue sections showed that the C.versicolor water extracts ameliorated liver injury in mice.
