儿童1型糖尿病外周血T细胞免疫球蛋白黏蛋白分子3和微小RNA-155的表达与糖代谢紊乱和微炎症状态的关系 被引量:4

Expression and relationship between Tim-3,miR-155 in peripheral blood and glucose metabolism disorders and microinflammatory states in children with type 1 diabetes mellitus disease

摘要 目的探讨儿童1型糖尿病(T1MD)外周血T细胞免疫球蛋白黏蛋白分子3(Tim-3)、微小RNA-155(miR-155)表达水平及其与糖代谢紊乱、微炎症状态的相关性。方法选取2016年1月-2019年6月住院治疗的T1MD患儿90例(T1DM组),同期体检健康儿童90例为对照组。检测并比较两组外周血Tim-3、miR-155、糖代谢指标[空腹血糖(FPG)、糖化血红蛋白(Hb A1c)、血清C肽(FC-P)]、炎症指标[白细胞介素-1β(IL-1β)、干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、转化生长因子-β(TGF-β)和白细胞介素-17(IL-17)]。结果T1DM组的外周血Tim-3、血清IL-1β、IFN-γ及IL-17水平高于对照组,miR-155、IL-4、IL-10及TGF-β水平低于对照组(P<0.05);T1DM患儿的miR-155与FPG、Hb A1c、FC-P、IL-1β、IFN-γ及IL-17呈负相关性,与IL-10、TGF-β呈正相关性(P<0.05);Tim-3与FPG、Hb A1c、FC-P、IL-1β、IFN-γ及IL-17呈正相关性,与miR-155、IL-4、IL-10及TGF-β呈负相关性(P<0.05)。结论儿童T1MD存在明显外周血Tim-3高表达和miR-155低表达,且与糖代谢紊乱和微炎症状态密切相关,可能作为儿童T1DM早期预测及诊治的潜在靶点。 Objective To investigate the expression and relationship between T cell immunoglobulin mucin molecule 3(Tim-3),micro RNA-155(miR-155)in peripheral blood and glucose metabolism disorders and microinflammatory states in children with type 1 diabetes mellitus disease(T1 MD).Methods From January 2016 to October 2019,90 cases of children with T1 MD who were hospitalized in our hospital as the T1 DM group,and 90 cases of healthy children who underwent physical examination during the same period were enrolled as the control group.The expression of Tim-3 and miR-155 of peripheral blood of the two groups were detected,the glucose metabolism indicators[fasting blood glucose(FPG),glycated hemoglobin(Hb A1 c),serum C peptide(FC-P)],inflammation indicators[interleukin-1β(IL-1β),interferon-γ(IFN-γ),interleukin-4(IL-4),interleukin-10(IL-10),transforming growth factor-β(TGF-β),interleukin-17(IL-17)]were detected and compared.Results The expression of peripheral blood Tim-3 and serum levels of IL-1β,IFN-γ,IL-17 in T1 DM group were higher than those in control group,and the expression of peripheral blood miR-155 and serum levels of IL-4,IL-10 and TGF-βwere lower than those in control group(P<0.05)..In children with T1 DM,the expression of peripheral blood miR-155 was negatively correlated with FPG,Hb A1 c,FC-P,IL-1β,IFN-γ,and IL-17,while it was positively correlated with IL-10 and TGF-β(P<0.05).;the expression of Tim-3 was positively correlated with FPG,Hb A1 c,FC-P,IL-1β,IFN-γ,IL-17,while it was negatively correlated with miR-155,IL-4,IL-10,TGF-β(P<0.05).Conclusion Children with T1 MD have obvious high expression of Tim-3 and low expression of miR-155 in peripheral blood.The expression of Tim-3 and miR-155 were closely related with glucose metabolism disorder and micro-inflammatory status in patients with T2 DM,which may be potential targets for early prediction,diagnosis and treatment for children with T1 DM.
