摘要
目的验证自行设计的热刺激联合Y迷宫实验用于观察糖尿病(DM)小鼠认知功能的可行性。方法腹腔注射链尿佐菌素130 mg/kg构建小鼠DM模型(DM组,12只),取14只小鼠为空白对照(BC组)。两组均进行自行设计的热刺激联合Y迷宫实验,比较两组建模后第6、8周热刺激联合Y迷宫实验的训练次数和错误次数、第7周Morris水迷宫实验逃避潜伏期及第9周空腹血糖和糖化血清蛋白水平。结果建模后DM组饮水量和进食量均大于BC组(P<0.01或P<0.05)。建模后第8周DM组错误次数多于BC组(P<0.01)。DM组第9、10次Morris水迷宫逃避潜伏期延长(P<0.05)。DM组糖化血清蛋白与BC组相仿[(5.14±3.48) mmol/L vs.(3.92±2.23) mmol/L](P>0.05)。DM组空腹血糖高于BC组[(17.23±9.61) mmol/L vs.(6.18±1.16) mmol/L](P<0.05)。结论自主设计热刺激联合Y迷宫实验用于观察DM小鼠认知功能具有可行性。
Objective To verify the feasibility of self-designed thermal stimulation combined with Y maze test in observing the cognitive function of the mice with diabetes mellitus(DM).MethodsDM model(group DM) was established in 12 mice by intraperitoneal injection of streptozotocin 130 mg/kg.Fourteen mice were taken as the blank controls(group BC).The self-designed thermal stimulation combined with Y maze test was performed in both groups.The indicators observed included the training times and error times of the thermal stimulation combined with Y maze test on the 6th and 8th week after modeling, the escape latency of morris water maze test on the 7th week after modeling, and the levels of fasting blood glucose and glycosylated serum protein on the 9th week after modeling, which were compared between the two groups.Results The amounts of water and food intake in group DM were more than those in group BC(P<0.01 or P<0.05).The number of errors in DM group was more than that in group BC on the 8th week after modeling(P<0.01).The escape latency at the 9th and 10th time test of morris water maze test was prolonged(P<0.05).The glycosylated serum protein of group DM was similar to that of group BC [(5.14±3.48) mmol/L vs.(3.92±2.23) mmol/L](P>0.05).Fasting blood glucose in group DM was higher than that in group BC [(17.23±9.61) mmol/L vs.(6.18±1.16) mmol/L](P<0.05).Conclusion The self-designed thermal stimulation combined with Y maze test is feasible in observing the cognitive function of the mice with DM.
