摘要
[目的]探究MAD2基因在文心兰‘柠檬绿’和拟南芥不同育性的株系中表达量的差异。[方法]通过对模式植物拟南芥的AtMAD2基因突变体的表型、结荚情况和花粉粒活力分析,结合已有研究结果综合分析拟南芥和文心兰中MAD2的表达差异。[结果]mad2突变体的抽梗时间早于野生型约4天。果荚长度单果荚种子数量都显著低于野生型植株,其中mad2突变体的果荚长度约为0.79 cm,单荚种子数约为7粒,野生型分别为1.07 cm、19粒。野生型拟南芥的花粉活力染色率为94%,mad2突变体的花粉活力染色率仅为69%,且拟南芥mad2突变体花粉粒部分发育异常。[结论]MAD2确实是影响花粉发育的关键基因之一,但在文心兰中育性与表达量同低,而在拟南芥中表达量增加会导致育性降低。
[Objective]To explore the difference of MAD2 gene expression in different fertility lines of Oncidium‘Honey Angel’and Arabidopsis thaliana.[Method]The phenotype,pod setting and pollen viability of the mutant of AtMAD2 gene in Arabidopsis thaliana were analyzed,and the differences of MAD2 expression between Arabidopsis thaliana and Oncidium were analyzed comprehensively.[Result]The results show that:(i)the stem time of mad2 mutant was about 4 days earlier than that of wild type.(ii)The number of seeds per pod in pod length was significantly lower than that in wild type.The length of pod of mad2 mutant is about 0.79 cm,and the number of seeds per pod is about 7.The wild type was 1.07 cm and 19.(iii)The pollen viability staining rate of wild type was 94%.The pollen viability staining rate of mad2 mutant was only 69%,and the pollen grain development of mad2 mutant was abnormal.[Conclusion]MAD2 is indeed one of the key genes affecting pollen development,but the fertility and expression are as low in Oncidium,while the increase in expression in Arabidopsis thaliana will lead to the decrease of fertility.
