环状RNA circ-MFN2在胰腺癌中的表达及其临床意义 被引量:1

Expression and clinical significance of circ-MFN2 in pancreatic cancer

摘要 目的探究环状RNA-线粒体融合蛋白2(circular RNA mitochondrial fusin 2,circ-MFN2)在胰腺癌中的表达,分析其与患者临床病理特征及预后间的相关性。方法采用qRT-PCR法检测55例胰腺癌患者的癌组织及血清中circ-MFN2 miRNA的表达,分析其与患者临床病理特征及预后间的相关性,计算circ-MFN2 miRNA表达诊断胰腺癌的敏感度、特异度和诊断准确率。根据ROC曲线分析其作为胰腺癌早期诊断生物标志物的效能。结果胰腺癌组织及血清中circ-MFN2 miRNA均呈高表达,分别与癌旁组织及健康对照组血清比较差异均具有统计学意义(均P<0.05)。胰腺癌组织中circ-MFN2 miRNA的表达与患者年龄、性别、肿瘤大小、病理学类型及肿瘤部位无关(P>0.05),但与CA19-9水平、TNM分期、肿瘤分化程度及淋巴结转移显著相关(P<0.05)。circMFN2 miRNA表达诊断胰腺癌的敏感度、特异度和准确率分别为72.7%、70.9%及83.6%,均分别高于CA19-9的54.5%、50.9%和52.7%(均P<0.05)。Kaplan-Meier分析显示circ-MFN2 miRNA高表达胰腺癌患者的中位生存时间显著短于低表达者(9.1个月比22.3个月,P<0.05)。血清circ-MFN2 miRNA作为诊断胰腺癌血清生物标志物的ROC曲线下面积为0.861[95%CI为(0.775,1.157),P=0.000]。Cox多因素分析显示,circ-MFN2 miRNA表达及淋巴结转移是胰腺癌患者预后的独立危险因素。结论circ-MFN2 miRNA在胰腺癌组织中呈高表达,且与患者临床病理特征及预后密切相关,有望成为预测胰腺癌预后的新型分子标志物。 Objective To investigate the expression of circular RNA mitochondrial fusin 2(circ-MFN2) in pancreatic cancer and analyze its correlation with clinicopathological features and prognosis. Methods The expressions of circ-MFN2 miRNA in 55 cases of pancreatic cancer tissues and serum were detected by qRT-PCR, and analyzed the correlation between circ-MFN2 and clinicopathological factors of pancreatic cancer and prognosis. The sensitivity,specificity and accuracy of expression of circ-MFN2 miRNA in the diagnosis of pancreatic cancer were statistically analyzed. ROC curve was used to analyze its efficacy as a biomarker for early diagnosis of pancreatic cancer.Results Compared with paracancerous tissues of pancreatic cancer and serum of healthy control group, circ-MFN2 miRNA was highly expressed in pancreatic cancer tissues and serum, and the difference were all statistically significant(all P<0.05). Chi square test showed that the expression of circ-MFN2 miRNA in pancreatic cancer tissues was not related to age, gender, tumor size, pathological type, and tumor site(P>0.05), but was significantly related to CA19-9 level, TNM stage, tumor differentiation and lymph node metastasis(P<0.05). The sensitivity, specificity and accuracy of expression of circ-MFN2 miRNA for pancreatic cancer were 72.7%, 70.9% and 83.6% respectively, which were all higher than that of CA19-9(54.5%, 50.9% and 52.7%, P<0.05). Kaplan-Meier analysis showed that the median survival time of pancreatic cancer patients with high expression of circ-MFN2 miRNA was significantly shorter than that of patients with low expression(9.1 months vs 22.3 months, P<0.05). The area under ROC curve of circ-MFN2 miRNA as a serum biomarker for the diagnosis of pancreatic cancer was 0.861 [95%CI(0.775, 1.157), P=0.000]. Cox multivariate analysis showed that the expression of cirC-MFN2 miRNA and lymph node metastasis were independent risk factors for the prognosis of pancreatic cancer patients. Conclusions Circ-MFN2 miRNA is highly expressed in pancreatic cancer tissues, and it is related to the clinical characteristics and prognosis of patients. It is expected to be a new molecular marker to predict the prognosis of pancreatic cancer.
