摘要
目的探究不同浓度艾烟对大鼠肺组织的毒性作用及其潜在的分子机制。方法将20只SD大鼠随机分为对照组、低浓度艾烟组、中浓度艾烟组、高浓度艾烟组;艾烟组大鼠分别放置于低、中、高浓度艾烟的染毒柜中,每天1 h,持续8周。比较各组大鼠肺灌洗液中SOD、TNF-α、IL-6和IL-10的表达情况;检测肺组织病理变化及NF-κB、TLR2和TLR4蛋白的表达情况。结果 (1)中低浓度艾烟组大鼠体质量显著高于对照组,而高浓度艾烟组大鼠体质量显著低于对照组(P<0.001)。(2)中高浓度艾烟组大鼠肺组织有明显的炎症细胞浸润,高浓度艾烟组大鼠肺组织发生纤维化;(3)中高浓度艾烟组大鼠肺组织中IL-6、TNF-α、NF-κB、TLR2和TLR4等炎症因子的表达显著增加,SOD和IL-10的表达降低(P<0.05)。结论低浓度艾烟对机体无显著性损害,高浓度艾烟引起的肺部损伤主要是通过促进炎症因子的表达产生的。
Objective To investigate the toxic effects of different concentrations of moxa smoke on rat lung tissues and to explore the potential molecular mechanisms. Methods Twenty SD rats were randomly divided into four groups: control group, low concentration group of moxa smoke, medium concentration group of moxa smoke and high concentration group of moxa smoke. Rats in the moxa smoke groups were respectively placed in the poisoning cabinet of low, medium and high concentration of moxa smoke for 1 hour per day for 8 weeks. The expressions of SOD, TNF-α, IL-6 and IL-10 in the lung lavage fluid of different groups were compared. The pathological changes of lung tissue and the expressions of NF-κB, TLR2 and TLR4 proteins were detected. Results(1)The body weight of rats in the medium and low concentration groups of moxa smoke was significantly higher than that of the control group, while the body weight of rats in the high concentration group of moxa smoke was significantly lower than that of the control group(P<0.001).(2)There were significantly inflammatory cell infiltration in the lung tissue of the medium and high concentration groups, and fibrosis occurred in the lung tissue of rats in the high concentration group.(3)The expressions of inflammatory factors such as IL-6, TNF-α, NF-κB, TLR2 and TLR4 in the rat lung tissue of the medium and high concentrations groups of moxa smoke were significantly increased, while the expressions of SOD and IL-10 were decreased(P<0.05). Conclusion Low concentrations of moxa smoke do not significantly damage the body, and the lung damage induced by high concentrations of moxa smoke is mainly produced by promoting the expression of inflammatory factors.
