摘要
目的研究黄连素(BBR)对人耐紫杉醇(PTX)去势性前列腺癌细胞凋亡和白细胞介素-6(IL-6)/信号转导与转录因子3(STAT3)表达的影响。方法选取对数生长期人耐PTX前列腺癌细胞株DU145 R,用DMSO、10及20μmol/L BBR处理72 h,采用流式细胞术检测DU145 R细胞凋亡;取0、10及20μmol/L BBR处理24 h的DU145 R细胞,采用实时荧光定量聚合酶链式反应(qPCR)与蛋白印迹(Western blot)分别检测DU145 R细胞中IL-6、STAT3 mRNA及蛋白表达。结果与DMSO组相比,10和20μmol/L BBR组DU145 R细胞凋亡数量增加(P<0.05);与0μmol/L组比较,10和20μmol/L BBR组DU145 R细胞中IL-6与STAT3 mRNA及蛋白表达降低(P<0.05)。结论BBR可能通过下调IL-6/STAT3表达诱导DU145 R细胞的凋亡。
Objective To study the effects of berberine(BBR)on apoptosis and interleukin-6(IL-6)/signal transducer and activator of transcription 3(STAT3)expression of paclitaxel(PTX)resistance in castration-resistant prostate cancer(CRPC)cells(DU145 R).Methods The logarithmic growth phase PTX-resistant prostate cancer cell strain DU145 R was selected and treated with DMSO,10 and 20μmol/L BBR for 72 h,and its apoptosis was detected by flow cytometry.After 24 hours of treatment with 0,10,and 20μmol/L of BBR respectively,the mRNA and protain expression of IL-6 and STAT3 in DU145 R cells of 3 groups were detected by quantitative real-time PCR(qPCR)and Western blot.Results Compared with the DMSO group,the number of apoptotic DU145 R cells were increased in the 10 and 20μmol/L BBR groups(P<0.05).Compared with the 0μmol/L group,the mRNA and protein expression of IL-6,STAT3 in DU145 R cells were decreased(P<0.05).Conclusion BBR induces apoptosis of DU145 R cells by down-regulating the expression of IL-6/STAT3.
