肝细胞癌组织表达下调蛋白的筛选及分析

Screening and analysis of protein with down-regulated expression in hepatocellular carcinoma

目的筛选肝细胞癌(HCC)组织表达下调蛋白,探索生物标志物。方法采用在中国人民解放军某三甲医院8例住院患者HCC组织及癌旁组织标本,使用同位素标记相对和绝对定量(iTRAQ)蛋白标记技术和液相串联质谱分析对这些标本进行蛋白表达水平研究。结果共筛选出下调蛋白71个,结合蛋白覆盖度、可信度及文献,发现蛋白谷胱甘肽转移酶(GSTM1)、B淋巴细胞瘤-2蛋白(Bcl-2)、细胞色素氧化酶(CYP)2C8、CYP2A6、氨基酰化酶1(ACY1)明显下调。结论HCC组织下调蛋白对该病的诊断、预后判断有重要价值,下调蛋白迫切需要大样本进行验证,并开展关键蛋白机制研究,为HCC的防治提供新的信息。

Objective Screening the protein with down-regulated expression in hepatocellular carcinoma(HCC)and explore the biomarkers.Methods HCC tissue and paracancer tissue specimens were collected from 8 inpatients in the People′s Liberation Army Hospital and the protein expression levels of these specimens were studied by isobaric tags for relative and absolute quantitation(iTRAQ)protein labeling and liquid phase tandem mass spectrometry.Results A total of 71 down-regulated proteins were screened,and it was found that glutathione S-transferase M1(GSTM1),B-cell lymphoma(Bcl-2),CYP2C8,CYP2A6 and Aminoacylase 1(ACY1)were significantly down-regulated in combination with protein coverage,reliability and literature.Conclusion Down-regulated proteins in HCC are of great value in the diagnosis and prognosis of the disease.It is urgent to verify the down-regulated proteins in large samples,and to carry out research on the mechanism of key proteins,so as to provide new information for the prevention and treatment of HCC.
