摘要
目的:探究类风湿关节炎患者外周血调节性T细胞(Treg细胞)分泌白细胞介素-35(IL-35)水平与其发病的相关性。方法:选取类风湿关节炎患者70例作为观察组,健康体检者70例为对照组。采集2组外周血,检测Treg细胞数量、Foxp3荧光强度、IL-35水平。同时将观察组患者按照28个关节疾病活动度评分(DAS28评分)分为不同的组,对各组Treg细胞、Foxp3荧光强度、IL-35水平进行检测。结果:观察组患者外周血中Treg细胞高于对照组,IL-35水平低于对照组,差异有统计学意义(P<0.05);2组Foxp3荧光强度比较,差异无统计学意义(P>0.05)。各DAS28评分组RA患者的Treg细胞、IL-35水平比较,差异有统计学意义(P<0.05),且DAS28评分越高,Treg细胞数量和IL-35水平越低;各DAS28评分组RA患者Foxp3荧光强度比较,差异无统计学意义(P>0.05)。结论:类风湿关节炎患者外周血中Treg细胞数量明显上升,IL-35水平显著下降。由于Treg细胞功能缺陷,降低了负调控能力。
Objective:To investigate the correlation between the level of IL-35 secreted by Treg cells in peripheral blood and the pathogenesis of rheumatoid arthritis(RA).Methods:Seventy RA patients were selected as an observation group and 70 healthy physical examinees were selected as a control group.The peripheral blood of both groups was collected.The number of Treg cells,the fluorescence intensity of Foxp3 and the level of IL-35 were measured.At the same time,the patients in the observation group were divided into different groups according to the DAS28 score,and the Treg cells,Foxp3 fluorescence intensity and IL-35 levels in each group were detected.Results:The level of Treg cells in peripheral blood of the observation group was higher than that of the control group,and the level of IL-35 was lower than that of the control group(P<0.05);There was no significant difference in the fluorescence intensity of Foxp3 between the two groups(P>0.05).The difference of Treg cells and IL-35 levels in RA patients with different DAS28 scores was statistically significant(P<0.05),and the higher the score of DAS28,the lower the number of Treg cells and IL-35 levels;There was no significant difference in the fluorescence intensity of Foxp3 between RA groups with different DAS28 scores(P>0.05).Conclusion:The number of Treg cells in the peripheral blood of RA patients increased significantly,and the level of IL-35 decreased significantly.Because of the deficiency of Treg cells,the negative regulation ability was reduced.
