东方拟无枝酸菌产万古霉素发酵工艺优化 被引量:2

摘要 采用东方拟无枝酸菌V19-07发酵产万古霉素,通过单因素实验对发酵工艺进行了优化,确定最优发酵工艺为:发酵温度28℃、发酵液pH值7.2、通气量1.0 vvm、溶解氧浓度10%。该工艺经50 L发酵罐验证,平均发酵效价达14117 U·mL^(-1),产量较优化前提高18.8%,可用于规模化生产。 Vancomycin was produced by fermentation of Amycolatopsis orientalis V19-07.We optimized the fermentation process by single-factor experiments.The optimum fermentation process is determined as follows:the fermentation temperature is 28℃,the pH value of fermentation broth is 7.2,the aeration rate is 1.0 vvm,and the dissolved oxygen concentration is 10%.In 50 L fermentor,the average fermentation titer of Vancomycin reaches 14117 U·mL^(-1) under the optimum process,while the yield increases by 18.8%compared with that under the original conditions,indicating that the process can be used for large-scale production.
