TUSC3基因表达对乳腺癌细胞迁移和侵袭能力的影响

摘要 目的本研究旨在探讨干扰乳腺癌细胞中TUSC3基因的表达后对乳腺癌细胞迁移和侵袭能力的影响。方法通过使用siRNA和过表达质粒分别建立TUSC3高/低表达的实验组,空质粒转染为对照组,采用划痕实验、Transwell实验检测各组乳腺癌细胞运动及侵袭能力的变化;采用western blot实验检测在乳腺癌细胞各实验组和对照组中E-cadherin、Vimentin的表达。结果与对照组比较,TUSC3高表达时,肿瘤细胞迁移能力、侵袭能力下降(P<0.05);TUSC3低表达时,肿瘤细胞运动能力、侵袭能力上升(P<0.05)。与对照组相比,抑制TUSC3表达时,E-cadherin表达下降,Vimentin表达上升(P<0.05);当TUSC3在细胞中过表达时,E-钙粘蛋白表达下降,未见统计学意义(P>0.05),波形蛋白下降(P<0.05)。结论TUSC3基因在乳腺癌细胞中高/低表达时抑制/增强了乳腺癌细胞的运动及侵袭能力,且其作用机制可能与EMT(上皮-间质转化)相关。 Objective To investigate the effect of interference with the expression of TUSC3 gene on the migration and invasion of breast cancer cells.Methods The high/low expression group of TUSC3 was established by siRNA and overexpression plasmid,respectively.The control group was transected with the empty plasmid.Scratch test and Transwell test were used to detect the movement and invasion ability of breast cancer cells in each group.The expressions of E-cadherin and vimentin were detected by Western blot.Results Compared with the control group,when the expression of TUSC3 was high,the motility and invasion ability of tumor cells were decreased(P<0.05);when the expression of TUSC3 was low,the motility and invasion ability of tumor cells were increased(P<0.05).Compared with the control group,the expression of E-cadherin decreased and the expression of vimentin increased(P<0.05);When TUSC3 was over-expressed in cells,the expression of E-cadherin decreased(P>0.05),but vimentin decreased(P<0.05).Conclusion High/low expression of TUSC3 gene in breast cancer cells inhibits/enhances the mobility and invasion of breast cancer cells,and its mechanism may be related to EMT(epithelial mesenchymal transition).
