目的探讨环状RNA-0001727(circRNA-0001727)在肝细胞癌组织中的表达及与预后的关系。方法选取2015年8月至2016年8月间在郑州大学第一附属医院接受治疗的118例肝细胞癌患者作为研究对象。采用荧光定量PCR(quantitative fluorescence PCR,qRT-PCR)法检测肝细胞癌患者hsa_circ_0001727的相对表达量,分析hsa_circ_0001727与肝细胞癌患者临床病理特征的关系,并分析影响肝细胞癌患者预后的因素。结果肝细胞癌组织hsa_circ_0001727相对表达量低于癌旁组织,差异有统计学意义(P<0.05)。hsa_circ_0001727与肝细胞癌患者BCLC分期、病灶数目、分化程度有关(P<0.05)。118例肝细胞癌患者三年生存率为68.64%。死亡组患者hsa_circ_0001727相对表达量低于生存组,差异有统计学意义(P<0.05)。hsa_circ_0001727评估肝细胞癌患者预后的ROC曲线下面积(Area Under Curve,AUC)、敏感度、特异性分别为0.827、81.08%、76.54%。Cox回归分析显示肝动脉化疗栓塞及hsa_circ_0001727与肝细胞癌患者预后密切相关。结论hsa_circ_0001727在肝细胞癌组织中低表达,检测hsa_circ_0001727有助于了解患者预后情况。

Objective To investigate the expression of circular RNA-0001727(hsa_circ_0001727)in hepatocellular carcinoma and its relationship with prognosis.Methods 118 patients with hepatocellular carcinoma who were treated in First Affiliated Hospital of Zhengzhou University from August 2015 to August 2016 were selected as the study subjects.Quantitative fluorescence PCR(qRT-PCR)was used to detect the relative expression of hsa_circ_0001727 in hepatocellular carcinoma patients,and its relationship with prognosis was analyzed.Results The relative expression of hsa_circ_0001727 in hepatocellular carcinoma tissues was lower than that in adjacent tissues,and the difference was statistically significant(P<0.05).Hsa_circ_0001727 was associated with BCLC staging,number of lesions,and differentiation in patients with hepatocellular carcinoma(P<0.05).The 3a survival rate of 118 patients with hepatocellular carcinoma was 68.64%.The relative expression of hsa_circ_0001727 in the death group was lower than that in the survival group,and the difference was statistically significant(P<0.05).The area under the ROC curve(AUC),sensitivity,and specificity of hsa_circ_0001727 for evaluating the prognosis of patients with hepatocellular carcinoma were 0.827,81.08%and 76.54%,respectively.Cox regression analysis showed that transcatheter arterial chemoembolization and hsa_circ_0001727 were closely related to the prognosis of patients with hepatocellular carcinoma.Conclusion hsa_circ_0001727 is lowly expressed in hepatocellular carcinoma tissues.Detecting the expression of hsa_circ_0001727 is helpful to understand the prognosis of patients.
