rhNGF滴眼液与α1-AT注射液联合应用对糖尿病视网膜病变模型大鼠的治疗作用及机制的研究

Therapeutic Effects and Mechanisms of rhNGF Eye Drops Combined with α1-AT Injection on Diabetic Retinopathy Model Rats

目的:通过动物体内法评价重组人神经细胞生长因子(rhNGF)滴眼液与α1-抗胰蛋白酶(α1-AT)注射液联合使用对糖尿病眼病模型大鼠的治疗作用及作用机制的研究;方法:制备大鼠糖尿病眼病模型,将成模的大鼠随机分5组——模型组、盐酸二甲双胍片组、rhNGF滴眼液组、rhNGF滴眼液+α1-AT注射液组、盐酸二甲双胍片+α1-AT注射液组,另设一正常对照组.每日给药1次,连续给药5周,给药结束后检测相关的血清生化指标、细胞因子、组织蛋白水平的变化及相关脏器的病理组织学检查;结果:rhNGF滴眼液组、rhNGF滴眼液+α1-AT注射液组对糖尿病眼病模型大鼠血清指标TNF-α、AngⅡ和视网膜的AGE、RAGE及其免疫调控因子P物质、ICAM-1、CD80等有一定的改善作用,但以rhNGF和α1-AT组合物组改善作用更显著.

Objective: to evaluate the therapeutic effect and mechanism of recombinant human nerve cell growth factor(rhNGF) eye drops combined with α1-AT(α1-antitrypsin) injection on diabetic eye disease model rats. Methods: A model of diabetic eye disease in rats was prepared, and the modeled rats were randomly divided into 5 groups: model group, metformin hydrochloride tablet group, rhNGF eye drops group, rhNGF eye drops + α1-AT injection group and a metformin hydrochloride tablets + α1-AT injection group, plus a normal control group, all with one dose a day for 5 consecutive weeks. Then the changes of serum biochemical indexes, cytokines, tissue protein levels and the pathological examination of related organs are collected. Results: rhNGF eye drops group and rhNGF eye drops + α1-AT injection group have a certain improvement on serum index TNF-α, AngⅡ and AGE, RAGE and immunoregulatory substance P, ICAM-1, CD80 of diabetic eye disease model rats. But the improvement effect is more significant in the combination of rhNGF and α1-AT.
