哮喘患儿血清miR-29b与外周血Th1、Th2细胞失衡的相关性研究 被引量:8

摘要 目的探究哮喘患儿血清微小核糖核酸-29b(miR-29b)与外周血辅助性T细胞亚型1/辅助性T细胞亚型2(Th1/Th2)细胞失衡相关性。方法选取2016年5月至2019年5月收治的支气管哮喘患儿104例为研究对象,根据患儿临床病情进展分为急性发作组51例,临床缓解组53例,另选取同期体检显示健康儿童48例为健康对照组。采用实时荧光定量(qRT-PCR)法检测血清miR-29b表达水平,流式细胞术检测外周血Th1、Th2细胞水平,酶联免疫吸附(ELISA)法检测血清白介素-2(IL-2)、IL-4、IL-5、IL-6、IL-10、干扰素-γ(IFN-γ)水平,分析miR-29b与Th1/Th2失衡相关性。结果健康对照组、临床缓解组、急性发作组外周血Th1、Th1/Th2及血清miR-29b、IL-2、IFN-γ水平依次显著降低(P<0.05),外周血Th2、嗜酸性粒细胞及血清总免疫球蛋白E(IgE)、IL-4、IL-5、IL-6、IL-10水平依次显著升高(P<0.05)。急性发作期及临床缓解期哮喘患儿血清miR-29b与外周血Th1/Th2水平均呈正相关(P<0.05)。哮喘患儿血清miR-29b与外周血嗜酸性粒细胞计数、血清总IgE、IL-4、IL-5、IL-6、IL-10水平呈负相关(P<0.05),与哮喘患儿肺功能指标1 s量(FEV1)、用力肺活量(FVC)、最高呼吸气流(PEF)水平及血清IL-2、IFN-γ水平呈正相关(P<0.05)。结论健康对照组、临床缓解组、急性发作组血清miR-29b水平依次显著降低,miR-29b低表达可能通过调控Th1/Th2比例失衡,影响相关炎性细胞因子变化,促进哮喘发生发展。 Objective To investigate the correlation between the imbalance of microRNA-29b(miR-29b)in serum and helper T cell subtype 1/helper T cell subtype 2(Th1/Th2)in peripheral blood in children patients with bronchial asthma.Methods A total of 104 children patients with bronchial asthma who were admitted and treated in our hospital from May 2016 to May 2019 were enrolled in the study.According to the clinical progress of the disease,the patients were divided into acute attack group(n=51)and clinical remission group(n=53),moreover,the other 48 healthy children were enrolled as control group.The expression levels of miR-29b in serum were detected by real-time fluorescence quantitative(qRT-PCR),and flow cytometry was used to detect the levels of Th1 and Th2 cells in peripheral blood,and the serum levels of IL-2,IL-4,IL-5,IL-6,IL-10 and IFN-γwere detected by ELISA.In addition the correlation between miR-29b and Th1/Th2 imbalance was analyzed.Results The levels of Th1,Th1/Th2 in peripheral blood,and serum levels of miR-29b,IL-2 and IFN-γwere significantly decreased in order in control group,clinical remission group and acute attack group(P<0.05),however,the levels of Th2,eosinophils in peripheral blood and the serum levels of total immunoglobulin E(IgE),IL-4,IL-5,IL-6,IL-10 were significantly increased in order of the three groups(P<0.05).There was a positive correlation between the levels of miR-29b in serum and those of Th1/Th2 in peripheral blood in acute attack group and clinical remission group(P<0.05).The serum levels miR-29b were negatively correlated with eosinophil count,and the serum levels of total IgE,IL-4,IL-5,IL-6 and IL-10 in patientts with bronchial asthma(P<0.05),however,which were positively correlated with forced expiratory volume in one second(FEV1),forced vital capacity(FVC),peak expiratory flow(PEF)and the serum levels of IL-2 and IFN-γ(P<0.05).Conclusion The serum levels of miR-29b are decreased in order in control group,clinical remission group and acute attack group,and the low expression of miR-29b may promote the pathogenesis and development of asthma by regulating the imbalance of Th1/Th2 ratio and influencing the changes of related inflammatory cytokines.
