三甲散加减对晚期喉癌患者病灶组织增殖、凋亡和侵袭相关蛋白表达的影响

Effects of Sanjia powder on the proliferation,apoptosis and expression of invasion related proteins of tumor tissues in patients with laryngeal carcinoma

摘要 目的研究三甲散加减对晚期喉癌患者病灶组织增殖、凋亡和侵袭相关蛋白表达的影响。方法选取2016年10月至2020年10月收治的70例晚期喉癌患者作为研究对象,随机分为对照组和联合组,每组35例。对照组接受常规放化疗,联合组在对照组基础上加用三甲散加减治疗,连续治疗8周。比较2组喉癌组织增殖相关蛋白(STAT3、CyclinD1、c-myc)、凋亡相关蛋白(Bax、Bcl-2、Caspase-3)、上皮间质化(EMT)相关蛋白(N-cadherin、Vimentin、E-cadherin)、侵袭相关蛋白(CD44v6、COX-2、MMP-9)及Wnt/β-catenin、PI3K/Akt信号通路关键蛋白表达。结果2组治疗前STAT3、CyclinD1、c-myc、Bax、Bcl-2、Caspase-3、N-cadherin、Vimentin、E-cadherin、CD44v6、COX-2、MMP-9、Wnt、β-catenin、PI3K、p-Akt蛋白表达比较差异无统计学意义(P>0.05);2组治疗后STAT3、CyclinD1、c-myc、Bcl-2、N-cadherin、Vimentin、CD44v6、COX-2、MMP-9、Wnt、β-catenin、PI3K、p-Akt蛋白表达均较治疗前显著降低,Bax、Caspase-3、E-cadherin蛋白表达均较治疗前显著升高,差异有统计学意义(P<0.05),且联合组治疗后STAT3、CyclinD1、c-myc、Bcl-2、N-cadherin、Vimentin、CD44v6、COX-2、MMP-9、Wnt、β-catenin、PI3K、p-Akt蛋白表达低于对照组,Bax、Caspase-3、E-cadherin蛋白表达高于对照组,差异有统计学意义(P<0.05)。结论在常规放化疗基础上联用三甲散加减治疗晚期喉癌通过调控相关蛋白表达抑制肿瘤细胞增殖、侵袭,并促进其凋亡,机制与抑制Wnt/β-catenin、PI3K/Akt信号通路活化有关。 Objective To investigate the effects of Sanjia powder on the proliferation,apoptosis and expression of invasion related proteins of tumor tissues in patients with laryngeal carcinoma.Methods A total of 70 patients with advanced laryngeal cancer who were treated in our hospital from October 2016 to October 2020 were enrolled in the study,who were randomly divided into control group(n=35)and combination treatment group(n=35).The patients in control group were treated by routine radiotherapy and chemotherapy,however,the patients in combination treatment group,on the basis of control group,were treated by Sanjia powder,with a treatment course of 8 weeks for both groups.The expression levels of proliferation related proteins(STAT3,CyclinD1 and c-myc),apoptosis related proteins(Bax,Bcl-2 and Caspase-3),EMT related proteins(N-cadherin,Vimentin and E-cadherin),invasion related proteins(CD44v6、COX-2,MMP-9),Wnt/β-catenin and PI3K/Akt pathway key proteins were observed and compared between the two groups.Results Before treatment,there were no significant differences in the expression levels of STAT3,CyclinD1,c-myc,Bax,Bcl-2,Caspase-3,N-cadherin,Vimentin,E-cadherin,CD44v6,COX-2,MMP-9,Wnt,β-catenin,PI3K and p-Akt between the two groups(P>0.05).After treatment,the expression levels of STAT3,CyclinD1,c-myc,Bcl-2,N-cadherin,Vimentin,CD44v6,COX-2,MMP-9,Wnt,β-catenin,PI3K and p-Akt in both groups were significantly decreased,however,the expression levels of Bax,Caspase-3 and E-cadherin were significantly increased,moreover,the increase degree and decrease degree in combination treatment group were more significant than those in control group(P<0.05).Conclusion Application of Sanjia powder on the basis of radiotherapy and chemotherapy can inhibit the proliferation and invasion,and promote the apoptosis of laryngeal cancer cells,and its action mechanism may be related to down-regulation of activation of Wnt/β-catenin and PI3K/Akt pathway.
