摘要
目的:探讨调节性T细胞(Treg)在梅毒血清固定形成中的作用。方法:分别选取30例梅毒血清固定患者、梅毒治疗后血清阴转患者及健康对照人群。采用流式细胞术检测CD4^(+)CD25^(+)叉状头螺旋转录因子(Foxp3)、Treg在CD4T细胞中的比例;采用实时荧光定量聚合酶链式反应(qPCR)检测外周血单个核细胞(PBMC)中Foxp3、细胞毒T淋巴细胞相关抗原(CTLA)-4、糖皮质激素诱导的肿瘤坏死因子受体(GITR)和转化生长因子(TGF)-βmRNA的表达;应用酶联免疫吸附试验(ELISA)检测血浆中CTLA-4、白细胞介素(IL)-10及TGF-β水平。结果:固定组、阴转组及健康对照组CD4*CD25^(+)Foxp3^(+)Treg在CD4^(+)T中的比例比较,差异有统计学意义(F=213.64,P<0.05),进一步两两比较示固定组比例均高于阴转组及健康对照组(P<0.05)。3组患者Foxp3、CTLA-4、GITR mRNA表达以及CTLA-4和IL-10含量比较,差异均有统计学意义(P<0.05);进--步两两比较示分别与阴转组及健康对照组比较,固定组表达均显著升高(P<0.05)。3组患者TGF-β含量及mRNA表达差异均无统计学意义(P>0.05)。结论:梅毒血清固定患者外周血CD4^(+)CD25Foxp3^(+)Treg数量增加,导致机体细胞免疫抑制,可能是造成梅毒血清固定的重要原因。
Objective:To investigate the potential roles of CD4^(+)CD25^(+)Foxp3^(+)Treg cells in serofast syphilis.Methods:Thirty patients with serorfast syphilis and 30 cases with cured syphilis were enrolled,and 30 healthy people were selected as healthy control group.The ratio of CD4CD25Foxp3^(+)Treg cells in CD4T cells was detect using flow cytometry.Quantitative real^(+)time PCR was used to detect the mRNA expression of Foxp3,CTLA-4,GITR and TGF-βin PBMC,and the serum levels of CTLA-4,IL-10 and TGF-βwas de-tected by ELISA.Results:The proportions of CD4^(+)CD25^(+)Foxp3^(+)Treg cells in serorfast syphilis group,seronegative syphilis group and healthy control group were significantly different(F=213.64,P<0.05),and the pairwise comparison showed that the proportion of CD4^(+)CD25^(+)Foxp3^(+)Treg in serofast syphilis group was significantly raised compared with seronegative syphilis group and healthy control group(P<0.05).The mRNA expressions of Foxp3,CTLA-4 and GITR,the contents of CTLA-4 and IL-10 in the three groups were also significantly different(P<0.05);the serofast syphilis group was significantly higher than seronegative syphilis group and healthy control group(P<0.05)according to pairvise comparison.There was no statistically significant difference in the content and mRNA expression of TGF-βamong the three groups(P>0.05).Conclusion:The increased number of CD4^(+)CD25^(+)Foxp3^(+)Treg cells in peripheral blood may induce cellular immunosuppression,which then triggers syphilis serofast.
