白藜芦醇通过调控Wnt/β-catenin信号通路抑制肝癌细胞生长的研究 被引量:5

摘要 目的探讨白藜芦醇(Res)对肝癌细胞Hep3B生长的影响及其与Wnt/β-catenin信号通路的关系。方法以25、50、100μmol/L的Res分别干预肝癌细胞株Hep3B 24、48、72 h后,用MTT法测定各组细胞增殖抑制率;用25、50、100μmol/L的Res干预Hep3B细胞24 h后,流式细胞术检测各组细胞周期变化情况;用Western blot法检测各组细胞Wnt、β-catenin、Lgr5的蛋白表达情况。结果25、50、100μmol/L的Res分别作用于Hep3B细胞24、48、72 h后,各组细胞的增殖速度均受到明显抑制,且抑制程度呈剂量与时间依赖性(P<0.05)。Res可阻断Hep3B细胞由G_(0)/G1期向S期的转化。不同浓度的Res作用于肝癌细胞株Hep3B 24 h后,其Wnt1、β-catenin、Lgr5的蛋白表达量均明显减少(P<0.05)。结论Res通过阻滞Hep3B细胞由G_(0)/G1期向S期的转化从而对其生长产生抑制作用,这可能是通过抑制Wnt/β-catenin信号通路的活化实现的。 Objective To investigate the influence of resveratrol(Res)on the growth of hepatoma carcinoma cells Hep3B and its relationship with Wnt/β-catenin signaling pathway.Methods Hepatocellular carcinoma cell line Hep3B was treated with 25,50 and 100μmol/L Res for 24,48 and 72 h,respectively,and cell proliferation inhibition rate in each group were measured by MTT assay;Hep3B cells were treated with 25,50,100μmol/L Res for 24 h,and the cell cycle changes in each group were detected by flow cytometry;the protein expressions of Wnt,β-catenin and Lgr5 in each group were detected by Western blot method.Results After Hep3B cells were treated with 25,50 and 100μmol/L Res for 24,48 and 72 h,respectively,the proliferation rate of cells in all groups was significantly inhibited with a dose-dependent and time-dependent manner(P<0.05).Res could block the transformation of Hep3B cells from G_(0)/G_(1) phase to S phase.After hepatocellular carcinoma cell line Hep3B were treated with Res at different concentrations for 24 h,the proteins expressions levels of Wnt1,β-catenin and Lgr5 in Hep3B cells significantly decreased(P<0.05).Conclusion RES inhibits the growth of Hep3B cells by blocking the transformation from G_(0)/G_(1) phase to S phase,which may be achieved by inhibiting the activation of the Wnt/β-catenin signaling pathway.
