摘要
目的:探讨不同浓度猪脱细胞真皮基质(pADM)对小鼠毛囊角质细胞迁移的诱导作用。方法:在Transwell实验和细胞划痕实验中,分别将0、2、10、20、40mgpADM配制成5个浓度梯度,分别作用于小鼠毛囊角质细胞。Transwell实验分组pADM浓度为0、4、20、40、80μg/μl;细胞划痕实验中pADM浓度分别为0、1、5、10、20μg/μl。每组重复3次。观察不同浓度pADM诱导毛囊角质细胞迁移的作用。结果:在Transwell实验中,0、4、20、40和80μg/μl组毛囊角质细胞计数分别为(51.67±2.96)、(129.78±2.91)、(187.11±0.84)、(159.89±2.69)和(161.44±3.4)个,差异具有统计学意义(P<0.05)。划痕实验中,0、1、5、10、20μg/μl组划痕愈合面积百分比分别为(1.8±0.3)%、(11.8±0.2)%、(32.4±0.6)%、(22.1±0.2)%和(21.8±0.2)%,差异具有统计学意义(P<0.05)。划痕愈合百分比随pADM浓度增加而逐渐增大,在5μg/μl组达到高峰,随后逐渐下降,其变化趋势与Transwell实验表现一致。结论:pADM具有诱导毛囊角质细胞迁移的作用,其作用大小与pADM浓度有关。
Objective:To investigate the effect of different concentrations of porcine acellular dermal matrix(pADM)on the migration of mouse hair follicle keratinocytes.Methods:In the Transwell experiment and the cell scratch experiment,pADM was formulated into 5 concentration gradients at 0,2,10,20,and 40 mg to act on mouse hair follicle keratinocytes respectively,and in the Transwell experiment,the cells were divided into five groups(0,4,20,40,80μg/μl groups)as per the pADM concentrations.In the cell scratch experiment,the cells was divided into five groups(0,1,5,10,20μg/μl groups)as per the pADM concentrations.The experiment was repeated 3 times.The migration ability of hair follicle keratinocytes induced by different concentrations of pADM was observed.Results:In Transwell test,the number of keratinocytes in hair follicles of 0,4,20,40 and 80μg/μl groups was(51.67±2.96),(129.78±2.91),(187.11±0.84),(159.89±2.69)and(161.44±3.4)respectively,indicating a statistically significant difference(P<0.05).In the scratch-wound healing assay,the percentage of scratch healing area in 0,1,5,10 and 20μg/μl groups was(1.8±0.3)%,(11.8±0.2)%,(32.4±0.6)%,(22.1±0.2)%and(21.8±0.2)%respectively,and the difference was statistically significant(P<0.05).The percentage of scratch healing increased gradually with the increase of pADM concentration,reached the peak in 5μg/μl group,and then decreased gradually,and its change trend was consistent with that of Transwell experiment.Conclusions:pADM can induce the migration of hair follicle keratinocytes,and its effect is related to the concentration of pADM.
