多指标层次分析法结合Box-Behnken响应面法优化连翘提取工艺 被引量:16

Optimization of extraction process of modified Forsythia suspensa by Box-Behnken response surface methodology plus multi-index analytic hierarchy process

摘要 目的:运用层次分析法(AHP)建立一种中药多指标的综合评价方法,优选连翘最佳提取工艺。方法:采用UPLC法同时测定连翘酯苷A、连翘苷和金丝桃苷的含量,层次分析法(AHP)计算权重系数,以多指标综合评分为评价指标,以醇体积分数、料液比和提取时间为影响因素,通过Box-Behnken响应面法优选连翘最佳提取工艺。结果:连翘最佳提取工艺为醇体积分数62.56%,料液比13.9,提取时间54.77 min。结论:优化工艺方法简便、预测性良好,结果可靠、评价指标可控,可用于规范连翘的提取工艺与质量控制。 OBJECTIVE To employ analytic hierarchy process(AHP)for establishing a comprehensive evaluation method of multiple indices of traditional Chinese medicine and optimizing the processing technology of Forsythia suspensa.METHODS The contents of forsythiaside A,phillyrin and hyperoside were simultaneously determined by ultra performance liquid chromatography(UPLC)and the method of AHP was utilized for calculating weight coefficient.The multi-index comprehensive score was utilized as an evaluation index while alcohol volume fraction,solid-liquid ratio and extraction time were selected as influencing factors.Extraction process of forsythia was optimized by Box-Behnken response surface method.RESULTS The optimal extraction process of forsythia was 62.56%methanol with a solid-liquid ratio of 13.9 and an extraction time of 54.77 min.CONCLUSION The above method offers the advantages of simple operation,excellent reproducibility and reliable outcomes.And this controllable evaluation index may be employed for quality control of forsythia.
