动脉球囊损伤大鼠Akt、GSK-3β表达及药物干预治疗

Expression of Akt and GSK-3β in rats with arterial balloon injury and drug intervention

摘要 目的观察糖代谢基因Akt、GSK-3β在动脉血管再狭窄中的表达及厄贝沙坦对其的影响。方法雄性Wistar大鼠48只,体重300~350 g,由沈阳医学院动物中心提供。制备大鼠动脉血管形成术模型,术后随机分为模型组、药物组。药物组(n=16):50 mg/(kg·d)厄贝沙坦溶于1 ml生理盐水,用灌胃针缓慢喂药;模型组(n=16)及对照组(n=16):等量生理盐水灌胃。Western blot测定Akt、GSK3β蛋白水平表达。结果动脉球囊损伤术后血管组织的Akt表达量较对照组明显升高(P<0.05),GSK3β表达量较对照组明显降低(P<0.05)。厄贝沙坦干预后Akt表达量较未干预组明显降低(P<0.05),GSK3β表达量较未干预组明显升高(P<0.05)。结论大鼠动脉球囊损伤术后血管组织中Akt表达升高,GSK3β表达降低,厄贝沙坦抑制动脉内膜增生可能与激活Akt、GSK-3β有关。 Objective To observe the expression of glucose metabolism genes Akt and GSK-3βin arterial restenosis and the effect of angiotensin receptor inhibitor-irbesartan on them.Methods A total of 48 male Wistar rats weighing 300~350 g were provided by Animal Center of Shenyang Medical College.The rat model of angioplasty was prepared.The rats were randomly divided into model group(n=16),drug group(n=16)and control group(n=16).The drug group was given irbesartan 50 mg/(kg·d)dissolved in 1 ml of normal saline,which was slowly administered with intragastric needle.The model group and control group were given the same amount of normal saline.Western blot was used to detect the expression of Akt and GSK-3β.Results The expression of Akt in vascular tissue of balloon injury group was significantly higher than that of control group(P<0.05),while the expression of GSK3βwas lower than that of control group(P<0.05).After irbesartan intervention,the expression of Akt in irbesartan group was significantly lower than that in non-intervention group(P<0.05),and the expression of GSK3βwas significantly higher than that in non-intervention group(P<0.05).Conclusion The expression of Akt is increased and the expression of GSK-3βis decreased after balloon injury.The inhibition of intimal hyperplasia by irbesartan may be related to the activation of Akt and GSK-3β.
