摘要
目的探讨超声造影定量达峰时间(TTP)、曲线下面积(AUC)水平与肝细胞癌细胞增殖的相关性。方法前瞻性抽取2020年1月至2020年12月商丘市立医院收治的76例肝细胞癌患者作为研究对象。对患者进行超声造影定量分析,获得相应的超声定量参数,包括TTP、平均渡越时间(mTT)、峰值强度(Imax)和AUC。采用实时荧光定量聚合酶链式反应(PCR)法检测肝癌细胞增殖相关基因的表达,包括畸胎癌衍生生长因子1(CR-1)、婆罗双树样基因4(SALL4)和促红细胞生成素肝细胞激酶(Eph)B4。分析超声造影定量参数水平与肝细胞癌细胞增殖相关基因表达的相关性。结果癌组织TTP、mTT低于癌旁组织,Imax、AUC高于癌旁组织,差异有统计学意义(P<0.05);癌组织CR-1、SALL4、EphB4表达高于癌旁组织,差异有统计学意义(P<0.05)。超声定量TTP、mTT与肝细胞癌细胞增殖相关基因CR-1、SALL4、EphB4表达呈负相关(r<0,P<0.05),Imax、AUC与肝细胞癌细胞增殖相关基因CR-1、SALL4、EphB4表达呈正相关(r>0,P<0.05)。结论超声造影定量TTP、mTT、Imax、AUC水平与肝细胞癌细胞增殖相关,随着超声定量TTP、mTT缩短,Imax、AUC升高,肝细胞癌增殖表达增加,超声造影定量参数可在一定程度上反映肝细胞癌癌细胞的增殖情况,为后续治疗提供指导。
Objective To investigate the correlation of quantitative time to peak(TTP)and area under curve(AUC)of contrast-enhanced ultrasound with the proliferation of hepatocellular carcinoma cells.Methods A total of 76 patients with hepatocellular carcinoma treated in Shangqiu Municipal Hospital from January 2020 to December 2020 were prospectively selected as the research subjects.Quantitative analysis of contrast-enhanced ultrasound of patients was performed,and the corresponding ultrasonic quantitative parameters were obtained,including TTP,mean transit time(mTT),peak intensity(Imax)and AUC.Real time fluorescent quantitative polymerase chain reaction(PCR)was used to detect the expression of proliferation related genes of hepatocellular carcinoma cells,including Cripto-1(CR-1),borneo double tree like gene 4(SALL4)and erythropoietin hepatocyte kinase(Eph)B4.The correlation of quantitative parameters of contrast-enhanced ultrasound with expression of proliferation related genes of hepatocellular carcinoma cells was analyzed.Results TTP and mTT in cancer tissues were lower than those in paracancerous tissues,while Imax and AUC were higher than those in paracancerous tissues(P<0.05).The expression of CR-1,SALL4,and EphB4 in cancer tissues were significantly higher than those in paracancerous tissues(P<0.05).The expression of CR-1,SALL4,and EphB4 of proliferation related genes of hepatocellular carcinoma cells was negatively correlated with quantitative TTP and mTT of contrast-enhanced ultrasound(r<0,P<0.05).Imax and AUC were positively correlated with the expression of CR-1,SALL4 and EphB4 of proliferation related genes of hepatocellular carcinoma cells(r>0,P<0.05).Conclusions The quantitative levels of TTP,mTT,Imax,and AUC of contrast-enhanced ultrasound are related to the proliferation of hepatocellular carcinoma cells.With the shortening of TTP and mTT,Imax and AUC are increased,and the expression of proliferation of hepatocellular carcinoma cell are increased.Quantitative parameters of contrast-enhanced ultrasound can reflect the proliferation of hepatocellular carcinoma cells in some degrees,and provide guidance for following treatment.
