平喘颗粒对哮喘气道相关炎症因子及肺组织HMGB1、TLR4表达的影响 被引量:6

Effects of Pingchuan Granules on Airway Related Inflammatory Factors and Expressions of HMGB1 and TLR4 in Lung Tissue of Asthma

摘要 目的观察平喘颗粒对哮喘气道相关炎症因子及肺组织高迁移率族蛋白B1(HMGB1)、Toll样受体4(TLR4)表达的影响。方法选择40只BALB/c小鼠随机分为空白组、模型组、布地奈德组、平喘颗粒组,建立哮喘模型,在末次激发24 h后取肺泡灌洗液(BALF)及肺组织标本。HE染色法观察小鼠肺组织炎症浸润情况及炎症评分情况;ELISA法检测小鼠BALF中白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平,同时对BALF中的嗜酸性粒细胞(EOS)及中性粒细胞(NEU)数量进行计数;Western blotting检测肺组织HMGB1、TLR4蛋白的表达。结果与空白组比较,模型组的HE染色情况及炎症评分升高,BALF中IL-1β、IL-6、TNF-α水平升高,BALF中EOS及NEU计数增加,肺组织HMGB1、TLR4表达升高(P<0.01);与模型组比较,布地奈德组和平喘颗粒组均可显著降低HE染色情况及炎症评分,降低BALF中IL-1β、IL-6、TNF-α的水平,减少BALF中EOS及NEU计数,降低肺组织HMGB1、TLR4表达(P<0.01)。结论平喘颗粒可有效减轻炎症细胞的浸润和相关炎症因子的水平,抑制HMGB1、TLR4蛋白表达,改善气道炎症,从而起到治疗哮喘的作用。 Objective:To observe the effect of Pingchuan Granules on airway related inflammatory factors and expressions of HMGB1 and TLR4 in lung tissue of asthma.Methods:Forty BALB/c mice were randomly divided into blank group,model group,budesonide group and Pingchuan Granules group to establish asthma model.BALF and lung tissue samples were collected 24 hours after the last challenge.HE staining was used to observe the in⁃flammatory infiltration and inflammatory score of lung tissue;ELISA was used to detect the levels of IL-1β,IL-6 and TNF-αin BALF,and the number of eosinophils(EOS)and neutrophils(NEU)in BALF was counted;Western blot was used to detect the expression of HMGB1 and TLR4 Protein in lung tissue.Results:Compared with the blank group,the HE staining and inflammation score,IL-1β,IL-6 and TNF-αlevels in BALF,EOS and NEU counts in BALF,and expressions of HMGB1 and TLR4 in lung tissue increased in the model group(P<0.01);compared with the model group,budesonide group and Pingchuan Granules group could significantly reduce HE staining and inflammation score,and the IL-1β,IL-6 and TNF-αlevels in BALF,the number of EOS and NEU in BALF was decreased,and the expression of HMGB1 and TLR4 in lung tissue was decreased(P<0.01).Conclu⁃sion:Pingchuan Granules can effectively reduce the infiltration of inflammatory cells and the level of related in⁃flammatory factors,inhibit the expression of HMGB1 and TLR4 Protein,improve airway inflammation,and play a role in the treatment of asthma.
