摘要
目的研究半夏泻心汤对脓毒症大鼠肠道损伤的影响,并基于HO-1的表达探讨半夏泻心汤减轻肠道损伤的作用机制。方法将60只雄性健康SD大鼠随机分为假手术组、脓毒症组、半夏泻心汤组。采用盲肠结扎穿孔法制备脓毒症大鼠模型,术后均予以生理盐水按10 mL/kg灌胃,半夏泻心汤组予半夏泻心汤10 mL/kg灌胃。造模后24 h麻醉取血并留取小肠标本。利用酶联免疫吸附试验(ELISA)法检测各组大鼠血清、肠道血红素加氧酶-1(HO-1)及肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)等炎症细胞因子的表达水平,同时进行大鼠肠道组织形态学检查,并观察小肠组织光镜下的病理变化及病理评分比较。结果脓毒症组及半夏泻心汤组大鼠血清及肠道IL-6、TNF-α含量较假手术组均明显升高,其中脓毒症组明显高于半夏泻心汤组(P<0.01);脓毒症组与半夏泻心汤组血清及肠道HO-1、IL-10含量较假手术组均明显升高,其中半夏泻心汤组明显高于脓毒症组(P<0.01);脓毒症组与半夏泻心汤组小肠组织病理学评分较假手术组明显升高,脓毒症组明显高于半夏泻心汤组(P<0.01)。结论半夏泻心汤能有效改善胃肠功能,对肠道有保护作用,其机制可能是半夏泻心汤通过上调HO-1表达,减轻炎症反应。
Objective:To study the effect of Banxia Xiexin Decoction on intestinal injury in septic rats and to explore the mechanism based on HO-1 expression.Methods:A total of 60 Sprague-Dawley(SD)male rats were randomly divided into sham group,sepsis group and Banxia Xiexin Decoction group.Septic rat model was estab⁃lished using cecal ligation puncture(CLP).Both the sham group and the sepsis group were given 10 mL/(kg·d)of normal saline,while Banxia Xiexin Decoction group were given 10 mL/(kg·d)of traditional Chinese medicine de⁃coction.Blood and ileum were collected 24 h after modeling.The concentration of HO-1,TNF-α,IL-6 and IL-10 in serum and ileum homogenate was measured by ELISA.Ileum was investigated by means of histopathology and pathological score of each group were compared.Results:The serum and intestinal IL-6 and TNF-αlevels of rats in the sepsis group and the Banxia Xiexin Decoction group were significantly higher than those in the sham group,and the sepsis group was significantly higher than that in the Banxia Xiexin Decoction group(P<0.01);the serum and intestinal HO-1 and IL-10 levels in the sepsis group and the Banxia Xiexin Decoction group were sig⁃nificantly higher than those in the sham group,and the Banxia Xiexin Decoction group was significantly higher than that in the sepsis group(P<0.01);the histopathological scores of the intestine in the sepsis group and the Banxia Xiexin Decoction group were significantly higher than those in the sham group,and the sepsis group was significantly higher than that in the Banxia Xiexin Decoction group(P<0.01).Conclusion:Banxia Xiexin Decoc⁃tion can alleviate intestinal injury in septic rats,and its mechanism may be related to up-regulation of HO-1 and reduction of inflammatory response.
