基于网络药理学探讨犀角地黄汤治疗过敏性紫癜的作用机制 被引量:5

Study on the Mechanism of Xijiao Dihuang Decoction in the Treatment of Henoch-Schonlein Purpura Based on Network Pharmacology

摘要 目的应用网络药理学方法探讨犀角地黄汤治疗过敏性紫癜的物质基础及作用机制。方法通过TC⁃MSP、ETCM、TCMID等在线数据库筛选水牛角、生地黄、牡丹皮、赤芍的有效成分及靶点预测,从OMIM与Gene⁃Cards数据库获取过敏性紫癜的相关疾病靶点,将药物靶点与疾病靶点进行交集,筛选得到共同作用靶点。将共同作用靶点利用String和Cytoscape软件进行蛋白互作网络构建及关键靶点筛选,并进行GO生物功能富集分析和KEGG通路富集分析。结果本研究共筛选得符合标准活性成分24个,药物潜在作用靶点452个,过敏性紫癜疾病靶点63个,取交集后获得药物与疾病共有核心靶点11个,GO富集分析得到20条相关条目,主要涉及炎症反应、氧化应激反应、信号传导、细胞凋亡等生物过程;KEGG富集分析筛选出12条通路,主要包括核心因子-κB信号通路、肿瘤坏死因子信号通路、癌症通路、NOD样受体信号通路、类风湿性关节炎、细胞黏附分子等。结论本研究构建了犀角地黄汤治疗过敏性紫癜多成分、多靶点、多通路的复杂网络关系,为进一步实验研究和临床运用提供了新的思路及参考依据。 Objective:To explore the material basis and mechanism of Xijiao Dihuang Decoction for treating Henoch-Schonlein purpura by the method of network pharmacology.Methods:The active ingredients and target prediction of Xijiao Dihuang Decoction were screened through online databases such as TCMSP,ETCM and TCMID.The related disease targets of Henoch-Schonlein purpura were obtained from the OMIM and GeneCards databases;the drug targets and disease targets were intersected and screened for common targets.String and Cyto⁃scape software was used to construct protein interaction network and screen key targets for common targets,and GO biological function enrichment analysis and KEGG pathway enrichment analysis were performed.Results:A total of 24 active ingredients,452 potential drug action targets and 63 targets of Henoch-Schonlein purpura dis⁃ease were screened,and 11 core targets of both drugs and diseases were obtained after intersection.Go enrichment analysis obtained 20 related items,mainly involving inflammatory response,oxidative stress response,signal trans⁃duction,cell apoptosis and other biological processes.Twelve pathways were screened out by KEGG enrichment analysis,mainly including NF-kappa B signaling pathway,TNF signaling pathway,Pathways in cancer,NOD-like receptor signaling pathway,Rheumatoid arthritis,Cell adhesion molecules,etc.Conclusion:In this study,the complex network relationship of multi-component,multi-target and multi-pathway of Xijiao Dihuang Decoction in the treatment of Henoch-Schonlein purpura was established,which has provided a new idea and reference for fur⁃ther experimental research and clinical application.
