基于网络药理学探讨苍附导痰丸治疗多囊卵巢综合征的作用机制 被引量:10

摘要 目的:运用网络药理学方法探讨苍附导痰丸治疗多囊卵巢综合症(polycystic ovary syndrome,PCOS)的作用靶点及分子机制。方法:在中药系统药理学数据库(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)检索苍附导痰丸的主要活性成分及对应的作用靶点;利用GeneCards数据库和OMIM数据库搜索多囊卵巢综合症的疾病靶点;运用R数据包图绘制苍附导痰丸作用靶点和多囊卵巢综合症的疾病靶点的交集,得到交集基因;通过String在线数据库构建蛋白互作网络;利用Cytoscape(3.2.1)构建“主要成分-疾病-靶点”网络图,分析其中的重要节点;通过R语言下的org数据包对得到的交集基因进行KEGG和GO富集分析。结果:网络药理学分析表明,共筛选出151个有效活性成分涉及通路,其中作用于PCOS的140个,通过105个潜在靶点发挥疗效。富集得到1900条GO生物信号,143条KEGG信号,筛选出前14个药物靶点。结论:苍附导痰丸可能通过调控JUN、MAPK3、HSP90AA1、MAPK1、ESR1、NR3C1、APP、MAPK14、MAPK8、VEGFA、EGFR、RB1、AR、IL6关键靶点,介导基因转录调节、脂质代谢、炎性微环境、胰岛素抵抗、性激素轴、动脉硬化、白介素免疫调节等信号,调控药物反应、营养与脂质代谢、活性氧、糖代谢、类固醇激素等生物进程调控达到治疗多囊卵巢综合症的作用。 Objective:To explore the target and molecular mechanism of Cangfu Daotan pill in the treatment of polycystic ovary syndrome(PCOS)by network pharmacology.Methods:The main active components and corresponding targets of Cangfu Daotan pills were searched in the database of traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP).GeneCards database and OMIM database were used to search for disease targets of PCOS;the intersection of the tar-get of Cangfu Daotan pill and the disease target of PCOS was drawn by using R data packet diagram to obtain the intersection gene;protein interaction network was constructed through String online database;Cytoscape(3.2.1)was used to construct a“ma-jor component-disease-target”network diagram and analyze the important nodes therein;KEGG and GO enrichment analyses were carried out on the obtained intersection genes through org data packets in R language.Results:Network pharmacology analy-sis showed that 151 active ingredients involved in pathways were screened out,among which acted on PCOS,and 105 potential targets were used to exert therapeutic effects.After enrichment,1900 GO biological signals and 143 KEGG signals were ob-tained,and the first 14 drug targets were screened out.Conclusion:Cangfu Daotan pill may mediate gene transcription regulation,lipid metabolism,inflammatory microenvironment,insulin resistance,sex hormone axis,arteriosclerosis,interleukin immune reg-ulation and other signals by regulating key targets of JUN,MAPK3,HSP90AA1,MAPK1,ESR1,NR3C1,APP,MAPK14,MAPK8,VEGFA,EGFR,RB1,AR,and IL6,and regulating biological processes such as drug response,nutrition and lipid metabolism,reactive oxygen species,sugar metabolism,steroid hormone,etc.as a may of treating PCOS.
