手持式猪精子密度仪在牛精子密度测定中的应用

摘要 为建立牛精子密度实用测定方法,实现密度仪一机多用,降低购买成本,以多头牛精液为材料,用手持式猪精子密度仪和血细胞计数板为检测工具,0.9%NaCl为检测液,建立牛精液吸光度-精子密度标准曲线(函数),并进行验证。结果显示:以0.9%NaCl为检测液时,牛精液吸光度-精子密度呈三次函数关系,回归曲线方程为Y=0.403X^(3)-0.165X^(2)+0.533X+0.001(R^(2)=0.980),吸光度最佳范围是0.108~1.131,在此范围内血细胞计数板法与函数方程得到的精子密度差异不显著(P>0.05);以血细胞计数板法测定的精子密度为真实值,函数计算密度平均相对误差为9.5%。本研究为建立动物的高效经济精子密度检测方法提供了参考。 The purpose of this study was to establish an efficient and economic method for testing the sperm density of bull, so as to realize the multi-purpose of one hand-held sperm density analyzer and to reduce the purchase cost of animal sperm density analyzers. The semen of several bulls were used as test samples. The hand-held sperm density analyzer and the blood cell counting board were used as detection tools. The 0.9% NaCl was used as the diluent for detection. The standard curve(function) of bull semen absorbance and sperm density was established and verified. The results showed that the relationship between the bull semen absorbance and sperm density was cubic when 0.9% NaCl was used as diluent for detection. The regression curve equation was Y=0.403X^(3)-0.165X^(2)+ 0.533X+0.001(R^(2)=0.980) and the best range of absorbance for testing was 0.108-1.131.In this range, there was no significant difference between the sperm density obtained using the blood cell counting plate method and the function equation(P>0.05). The average relative error was 9.5% when the sperm density measured by blood cell counting plate method was used as the real value. This study provided a reference for the establishment of an efficient economic method for sperm density detection in animals.
