摘要
为研究牛磺酸(taurine)对脂多糖(lipopolysaccharide,LPS)诱导的奶牛肝脏原代细胞(BHEC)炎症反应的保护作用及其相关信号通路的调控,试验分为4组,对照组(CON)、牛磺酸组(TAU)、LPS组(LPS)、牛磺酸+LPS组(LT),分别对炎性因子白介素-1β(IL-1β)、白介素-6(IL-6)、白介素-8(IL-8)和核因子κB亚基P65(P65)、核因子κB抑制蛋白(IκB)、Toll样受体4(TLR4)的mRNA水平,磷酸化核因子κB亚基P65(p-P65)、P65、磷酸化核因子κB抑制蛋白(p-IκB)、IκB、TLR4的蛋白水平以及P65在细胞中的定位进行测定。结果显示:与对照组相比,LPS组IL-1β、IL-6、IL-8、P65、TLR4的mRNA表达量显著升高(P<0.05),p-P65、p-IκB/IκB、TLR4的蛋白水平显著升高(P<0.05),P65蛋白由胞浆移入胞核;与LPS组相比,LT组IL-1β、IL-6、IL-8、P65、TLR4的mRNA表达量显著降低(P<0.05),p-P65、p-IκB/IκB、TLR4的蛋白水平显著降低(P<0.05),P65蛋白由胞核转移入胞浆。结果表明:牛磺酸预处理能显著降低LPS引起的BHEC中IL-1β、IL-6、IL-8、P65、TLR4的mRNA水平和p-P65、p-IκB/IκB、TLR4的蛋白表达,抑制P65入核,从而抑制LPS引起的BHEC炎症反应。
To determine the protective effect of taurine on primary cell inflammation in dairy cow liver induced by LPS,the present BHEC test consisted of four groups:the control group(CON group),the taurine treated group(TAU group),the LPS treated group(LPS group),the taurine and LPS treated group(LT group).The mRNA expression of IL-1β,IL-6,IL-8,P65,TLR4,IκBα,the protein expression of p-P65,P65,p-IκB,IκB,TLR4 and the location of P65 in the cell were measured.The results showed that,compared with the CON group,the expressions of IL-1β,IL-6,IL-8,P65,TLR4 mRNA and p-P65,p-IκB,TLR4 protein were significantly increased(P<0.05),and P65 entered the nucleus clearly.After taurine treatment,and compared with the LPS group,the expressions of IL-1β,IL-6,IL-8,P65,TLR4 mRNA and p-P65,p-IκB,TLR4 protein were significantly decreased(P<0.05),and the intrusion of P65 into the nucleus was prohibited.In conclusion,our study indicated that taurine suppressed LPS-induced inflammation in BHEC.
