芜菁中性多糖对D-半乳糖致衰老小鼠的抗氧化作用 被引量:12

Antioxidant Effect of Turnip Neutral Polysaccharide on D-Galactose-Induced Aging Mice

摘要 目的:研究芜菁中性多糖(Brassica rapa L.neutral polysaccharides,BRNP)对D-半乳糖致衰老小鼠模型的抗氧化作用。方法:昆明小鼠随机分为空白组、模型组、VE对照组和BRNP高、中、低剂量组,连续灌胃42 d后摘眼球取血测定各组小鼠血清以及全脑、肝脏匀浆中的超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)、过氧化氢酶(CAT)、还原性谷脱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,采用HE染色观察肝组织病理学改变。结果:与空白组相比,模型组血清、全脑及肝脏匀浆中SOD、T-AOC、CAT、GSH、GSH-Px水平明显下降,而MDA水平显著上升;药物干预组与模型组相比,显著提高血清以及全脑、肝脏匀浆中的SOD、T-AOC、CAT、GSH、GSH-Px的活性,并显著降低MDA含量;HE染色发现BRNP对衰老小鼠肝脏组织具有一定的保护作用。结论:BRNP能有效提高D-半乳糖致衰老小鼠模型抗氧化能力,达到延缓衰老的作用。 Objective:To study the antioxidant effect of Brassica rapa L.neutral polysaccharides (BRNP) on D-galactose induced aging mouse model.Methods:Kunming mice were randomly divided into blank group,model group,VE control group and BRNP high-dose,medium-dose and low-dose groups.The activities of superoxide dismutase (SOD),total antioxidant capacity (T-AOC),catalase (CAT),reduced glutathione (GSH),glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) in serum,whole brain and liver homogenates of mice were determined after 42 days of continuous intragastric administration.The histopathological changes of liver were observed by HE staining.Results:Compared with blank group,the levels of SOD,T-AOC,CAT,GSH,GSH-Px in serum,whole brain and liver homogenate of model group were significantly decreased,while the level of MDA was significantly increased.Compared with the model group,the activities of SOD,T-AOC,CAT,GSH and GSH-Px in serum,whole brain and liver homogenate were significantly increased in the drug intervention group,and the content of MDA was significantly decreased.HE staining showed that BRNP had a certain protective effect on the liver tissue of aging mice.Conclusion:BRNP can effectively improve the antioxidant capacity of D-galactose induced aging mouse model and achieve the effect of delaying aging.
