涎腺良恶性病变组织中人乳头瘤病毒基因的检测分析

摘要 目的探讨涎腺良恶性疾病与人乳头瘤病毒(human papilloma virus,HPV)感染的相关性。方法采用HPV基因通用引物GP5+/GP6+和CPI/CPIIG应用聚合酶链反应(polymerase chain reaction,PCR)技术对33例涎腺恶性肿瘤、35例涎腺良性肿瘤及18例涎腺良性非肿瘤病变石蜡包埋组织进行HPV基因检测,PCR产物采用琼脂糖凝胶电泳与测序进行鉴定。通用引物PCR阳性样本进一步采用HPV16/18型特异性引物进行PCR检测。结果 86例涎腺良恶性病变组织中HPV基因总检出率为25.6%(22/86),其中恶性肿瘤为18.2%(6/33),良性肿瘤为25.7%(9/35),良性非肿瘤病变为38.9%(7/18),三者间比较,差异无统计学意义(χ^(2)=2.624,P=0.269)。HPV16和HPV18基因在上述阳性样本中均未检出。结论 3种涎腺良恶性病变组织中均存在HPV感染,但HPV感染可能并非涎腺恶性肿瘤的主要致病因素。 Objective To evaluate the correlation between benign and malignant lesions of salivary gland and HPV infection.Methods Polymerase chain reaction(PCR)with consensus primers GP5+/GP6+ and CPI/CPIIG was used to detect HPV DNA in 86 cases of formalin-fixed,paraffin-embedded salivary gland lesion tissues including 33 cases of malignant tumors,35 cases of benign tumors and 18 cases of non-neoplastic lesions.Type-specific PCR was further conducted to detect E6 gene of HPV16 and HPV18 in positive tissues of consensus PCR.Results The results showed that the total positive rate of HPV DNA was 25.6%(22/86),and it was 18.2%(6/33),25.7%(9/35)and 38.9%(7/18)for malignant tumors,benign tumors and non-neoplastic lesions,respectively.There was no statistical significance among these three groups(χ^(2)=2.624,P=0.269).HPV16 and HPV18 genes were not detected in the above positive samples.Conclusion HPV exists in different kinds of salivary gland lesion tissues,but it may not play a crucial role in carcinogenesis of malignant salivary gland tumors.
