五味子乙素对慢性酒精中毒大鼠海马突触相关蛋白表达的影响 被引量:7

Effect of Schisandrin B on the expressions of hippocampal synapses associated proteins in rats with chronic alcoholism

摘要 目的研究五味子乙素对慢性酒精中毒大鼠学习记忆能力及突触相关蛋白表达的影响。方法连续用白酒以灌胃的方式建立动物慢性酒精中毒模型,随机将中毒模型制备成功后的大鼠分为模型组和低、中、高剂量实验组,每组均8只;另取8只正常大鼠作为空白组。低、中、高剂量实验组分别按10,20,40mg·kg-1的剂量给予0.01 g·m L^(-1)五味子乙素;空白组和模型组均给予等体积蒸馏水。5组大鼠每天灌胃给药1次,连续给药30 d。用Morris水迷宫测试大鼠定位航行和空间探索的潜伏期,用Western Blot检测海马组织中突触素(SYN)和突触后致密蛋白95(PSD-95)蛋白的表达水平。结果低、高剂量实验组和模型组、空白组的定位航行潜伏期分别为(33.19±13.90),(24.41±9.29),(41.44±10.30)和(20.47±10.65)s,空间探索潜伏期分别为(33.44±16.53),(21.88±18.62),(40.23±14.93)和(16.88±11.27)s,海马组织中PSD-95蛋白相对表达量分别为0.67±0.18,1.25±0.38,0.56±0.21和1.73±0.33,海马组织中SYN蛋白相对表达量分别为0.74±0.29,1.12±0.45,0.44±0.30和1.04±0.23。高剂量实验组和空白组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论五味子乙素可能通过上调SYN和PSD-95的表达,从而提高慢性酒精中毒大鼠学习记忆能力。 Objective To investigate the effects of Schizandrin B on the learning and memory ability and the expressions of synaptic proteins in rats with chronic alcoholism.Methods The animal model of chronic alcoholism was established by continuous gavage with liquor.The rats after successful modelling were randomly divided into model group and the experimental-L,-M,-H groups,with 8 rats in each group.Another 8 rats were selected as the blank group.The experimental-L,-M,-H groups were given 10,20 and 40 mg·kg-1 of 0.01 g·m L^(-1)Schizandrin B by gavage,respectively.The blank and model groups were given equal volumes of distilled water.Five groups were treated for 30days with once a day.Morris water maze was used to test the latency of positioning navigation and spatial exploration of rats.Western Blot was employed to detect the expressions of synaptophysin(SYN)and postsynaptic density protein-95(PSD-95)in the hippocampus of rats.Results The navigation latency of the experimental-L and-H groups,model group and blank group were(33.19±13.90),(24.41±9.29),(41.44±10.30)and(20.47±10.65)s,the latency of space exploration were(33.44±16.53),(21.88±18.62),(40.23±14.93)and(16.88±11.27)s,the relative expression levels of PSD-95 protein in hippocampus were 0.67±0.18,1.25±0.38,0.56±0.21 and 1.73±0.33,the relative expression levels of SYN proteins in hippocampus tissues were0.74±0.29,1.12±0.45,0.44±0.30 and 1.04±0.23,respectively.Compared with those of the model group,the above indexes of the high-dose experimental group and the blank group were significantly different(P<0.05,P<0.01).Conclusion Schisandrin B can improve the learning and memory ability of rats with chronic alcoholism,which may be associated with the upreguation of the protein expressions of SYN and PSD-95 in hippocampus.
