自噬相关基因Beclin1与乳腺癌病理相关性分析

摘要 目的探讨自噬相关基因Beclin1在乳腺癌中的表达情况和与乳腺癌相关病理的相关性。方法选取120例乳腺癌患者作为研究对象,将其分为恶性组,所有患者均行手术或空心针穿刺病理证实为乳腺癌患者,取同期120例良性乳腺肿瘤患者作为良性组,另取同期120例健康者的乳腺组织进行分析,将其分为对照组。分析三组患者自噬相关基因Beclin1的表达情况及阳性率,120例乳腺癌患者的临床病理情况,并应用回归分析,分析自噬相关基因Beclin1与乳腺癌病理相关性。结果通过对比三组中自噬相关基因Beclin1mRNA表达水平发现,良性组和恶性组均显著低于对照组,且恶性组显著低于良性组,对比差异显著(P<0.05);通过对比三组自噬相关基因Beclin1蛋白表达阳性率发现,良性组阳性率为60.00%,恶性组患者阳性率为26.67%,对照组阳性率为91.67%,对比差异显著(P<0.05);120例乳腺癌患者中,Beclin1阳性与Beclin1阴性患者月经状态、年龄、肿瘤大小对比无明显差异(P>0.05),两组患者的临床分期、淋巴结转移、组织分化程度对比差异显著(P<0.05);Spearman相关分析显示,月经状态、年龄、肿瘤大小与自噬相关基因Beclin1不相关(P>0.05),临床分期、淋巴结转移和组织分化程度与自噬相关基因Beclin1呈负相关(P<0.05)。结论自噬相关基因Beclin1在乳腺癌患者的癌组织中表达低于良性肿瘤患者和健康人士,随着患者病情加重,自噬相关基因Beclin1表达明显降低,Beclin1与乳腺癌患者的临床分期、淋巴结转移和组织分化程度呈负相关,因此可以应用自噬相关基因Beclin1来判断乳腺癌患者病情的严重程度。 Objective To investigate the expression of autophagy related gene Beclin1 in breast cancer and its correlation with breast cancer related pathology.Methods 120 cases of breast cancer patients were selected as the research object,and they were divided into malignant group.All patients were confirmed as breast cancer patients by surgery or hollow needle biopsy pathology.120 cases of benign breast tumor patients were selected as benign group,and the breast tissue of 120 healthy people in the same period were analyzed and divided into the control group.The expression and positive rate of autophagy related gene Beclin1 in 3 groups were analyzed.The clinicopathological features of 120 patients with breast cancer were analyzed.The correlation between Beclin1 and breast cancer pathology was analyzed by regression analysis.Results By comparing the 3 groups of autophagy related gene Beclin1 mRNA expression level,both the benign group and the malignant group were significantly lower than the control group,and the malignant group was significantly lower than the benign group,the difference was significant(P<0.05);by comparing the positive rate of the 3 groups of autophagy related gene Beclin1 protein expression,the positive rate of benign group was 60.00%,the positive rate of malignant group was 26.67%,and the positive rate of the control group was 91.67%There were no significant differences in menstrual status,age and tumor size between Beclin1 positive and Beclin1 negative patients(P>0.05),but there were significant differences in clinical stage,lymph node metastasis and tissue differentiation between the 2 groups(P<0.05).Spearman correlation analysis showed that menstrual status,age,tumor size and autophagy related gene BEC were correlated with menstrual status,age,tumor size There was no correlation between lin1 and autophagy related gene Beclin1(P>0.05).Clinical stage,lymph node metastasis and tissue differentiation were negatively correlated with Beclin1(P<0.05).Conclusion The expression of autophagy related gene Beclin1 in breast cancer patients is lower than that in benign tumor patients and healthy people.With the aggravation of patients'condition,the expression of autophagy related gene Beclin1 is signifi-cantly decreased.Beclin1 is negatively correlated with clinical stage,lymph node metastasis and tissue differentiation of breast cancer patients,so we can use Beclin1 to judge breast cancer patients The severity of the disease.
