菊粉对不同时期2型糖尿病小鼠维生素D的影响 被引量:3

摘要 目的探讨不同时期2型糖尿病小鼠在菊粉干预前后维生素D水平及其与炎症因子和肠道菌群的相关性。方法采用4周龄自发性2型糖尿病雌性小鼠(db/db)为模型,共40只。行糖耐量试验,分为糖尿病前期组(PDM组,20只)和糖尿病期组(DM组,20只),再从PDM组和DM组分别随机取出10只作为菊粉干预糖尿病前期组(INU/PDM组)和菊粉干预糖尿病期组(INU/DM组)。分别检测各组小鼠25(OH)D_(3)、IL-6、IL-10、TNF-α及LPS水平。结果菊粉干预INU/PDM及INU/DM组与未干预组相比,25(OH)D_(3)水平显著升高,差异有统计学意义(P<0.05)。IL-6、TNF-α及LPS与25(OH)D_(3)呈负相关性(r=-0.5324、-0.5401、-0.7772,P<0.05),与IL-10呈正相关性(r=0.5083,P<0.05);25(OH)D_(3)与产短链脂肪酸SCFAs的拟杆菌呈正相关性(r=0.7214,P<0.05),与黏螺菌和反刍螺菌_6呈负相关(r=-0.7394、-0.4685,P<0.05)。结论菊粉通过调节肠道菌群增加不同时期2型糖尿病,尤其是糖尿病前期小鼠的维生素D水平。调节肠道菌群可能成为早期治疗2型糖尿病伴低维生素D的新手段。 Objective To investigate the level of vitamin D and the correlation with inflammatory factors and intestinal flora in different stages of type 2 diabetes mice treated by inulin.Methods Forty 4-week-old female mice with spontaneous type 2 diabetes mellitus(db/db)were used as the model.According to the glucose tolerance tests,they were divided into the pre-diabetes group(PDM group,n=20)and the diabetes group(DM group,n=20).Ten mice were randomly selected from the PDM group and the DM group as inulin intervention of PDM group(INU/PDM group)and inulin intervention of DM group(INU/DM group),respectively.The level of 25-hydroxyvitamin D,IL-6,IL-10,TNF-a and LPS were detected in each group.Results The levels of 25-hydroxyvitamin D in INU/PDM and INU/DM groups were significantly increased than those in control group(P<0.05).Furthermore,Il-6,TNF-a and LPS were negatively correlated with 25(OH)D_(3)(r=-0.5324,-0.5401-0.7772,P<0.05)and positively correlated with IL-10(r=0.5083,P<0.05);25(OH)D_(3) was positively correlated with Bacteroides producing the short-chain fatty acid(SCFAs,R=0.7214,P<0.05),and negatively correlated with proinflammatory Mucispirillum and Ruminclostridium 6(r=-0.7394,-0.4685,P<0.05).Conclusion Dietary inulin can improve the level of vitamin D in diverse stages of T2DM via modulating gut microbiota,especially in pre-diabetic stages,thus regulation of intestinal flora may be a new method for early treatment of T2DM patients with low vitamin D.
