大叶千斤拔黄酮类成分提取及抗氧化活性研究 被引量:4

摘要 通过单因素试验及响应面试验优化大叶千斤拔总黄酮的乙醇回流提取工艺条件,并对提取物的抗氧化活性进行评价。结果表明:优化后的提取工艺条件为提取时间52 min,料液比(m大叶千斤拔∶V乙醇)1∶6(g/mL),乙醇体积分数70%,提取温度60℃,该条件下提取物的总黄酮含量为75.47μg/mg;提取物对ABTS+自由基、DPPH自由基清除能力的IC 50分别为0.0449,0.2123 mg/mL,总抗氧化能力及对Cu 2+和Fe 3+还原能力的IC 50分别为0.3896,0.2219,0.7317 mg/mL。说明建立的最佳提取工艺可有效获取大叶千斤拔总黄酮成分;大叶千斤拔黄酮类物质具有较强的体外抗氧化活性。 The extraction process conditions of total flavonoids from Flemingia macrophylla were optimized,and the extract's antioxidant properties were also studied.Firstly,the single factor test and response surface test were applied to optimize the alcohol reflux extraction process,and the antioxidant activity of the extract was finally evaluated.It was found that the best extraction time was 52 min,material-liquid ratio 1∶6(g/mL),ethanol volume fraction 70%,extraction temperature 60℃.Rutin was used as the standard substance,the total flavonoid content of the extract is 75.47μg/mg;the IC 50 of the extract for ABTS+·and DPPH·scavenging was 0.0449,0.2123 mg/mL,respectively,and the IC 50 for the total antioxidant capacity and the reducing ability of Cu 2+and Fe 3+is 0.3896,0.2219,0.7317 mg/mL.The best extraction process was established in this study,which could effectively obtain total flavonoids from Flemingia macrophylla;The total flavonoids extracted from Flemingia macrophylla showed intense in vitro antioxidant activity.
