“夹脊”电针对轴向加压致腰椎间盘退变模型兔IL-1β、TGF-β1表达的影响 被引量:4

摘要 目的研究"夹脊"电针对轴向加压致腰椎间盘退变模型兔IL-1β、TGF-β1的影响,探讨电针干预椎间盘退变的可能机制。方法将24只新西兰兔随机分为正常组(n=6)、假模型组(n=6)、模型组(n=6)、电针组(n=6)。采用L4、L5椎体间横穿入两根平行的克氏针并连接轴向加压造模器的方法,建立兔椎间盘退变模型,在验证造模成功后,电针组施以"夹脊"穴电针治疗28 d;模型组仅造模,不予针刺治疗;假模型组仅在L4、L5椎体间穿入克氏针,不安装轴向加压造模器,不做针刺治疗;正常组予以自然饲养,不做特殊处理。造模后第28天处死各组实验兔,取出椎间盘组织,运用免疫荧光及ELISA方法检测IL-1β、TGF-β1的表达情况。结果与正常组同期比较,假模型组实验兔椎间盘组织中IL-1β和TGF-β1的平均光密度及含量差异均无统计学意义(P>0.05);模型组实验兔椎间盘组织中IL-1β和TGF-β1的平均光密度及含量均明显增强,差异有统计学意义(P<0.01);经过"夹脊"电针干预后,电针组实验兔椎间盘组织中IL-1β和TGF-β1的平均光密度及含量均明显减弱,差异有统计学意义(P<0.01)。结论"夹脊"电针通过下调IL-1β、TGF-β1的表达,减少细胞凋亡,从而抑制和延缓腰椎间盘退变。 Objective To study the effects of"Jiaji"electroacupuncture on IL-1βand TGF-β1 in the rabbit model of lumbar intervertebral disc degeneration induced by axial compression,and to explore the possible mechanism of electroacupuncture intervention in intervertebral disc degeneration.Methods Twenty-four New Zealand rabbits were randomly divided into normal group(n=6),sham model group(n=6),model group(n=6),and electroacupuncture group(n=6).The rabbit intervertebral disc degeneration model was established by inserting two parallel Kirschner wires across between the L4 and L5 vertebral bodies and connecting the axial compression model.After verifying the success of the model,the electro-acupuncture group was applied with"Jianji"Acupoint electro-acupuncture treatment for 28 days.The model group only has a model,no acupuncture treatment.The sham model group only inserts Kirschner wires between the L4 and L5 vertebral bodies,and does not install the axial compression molder,and does not make needles Needle treatment.The normal group was fed naturally without special treatment.On the 28 th day after modeling,the experimental rabbits in each group were sacrificed,the intervertebral disc tissues were taken out,and the expression of IL-1βand TGF-β1 were detected by immunofluorescence and ELISA methods.Results Compared with the normal group at the same time,there was no significant difference in the average optical density and content of IL-1βand TGF-β1 in the experimental rabbit intervertebral disc tissue of the sham model group(P>0.05).IL-1βin the experimental rabbit intervertebral disc tissue of the model group The average optical density and content of 1βand TGF-β1 were significantly enhanced(P<0.01).After the intervention of"Jiaji"electroacupuncture,the average optical density of IL-1βand TGF-β1 in the intervertebral disc tissue of experimental rabbits in the electroacupuncture group and The content was significantly weakened(P<0.01).Conclusion"Jiaji"electroacupuncture can inhibit and delay the degeneration of lumbar intervertebral discs by down-regulating the expression of IL-1βand TGF-β1,reducing cell apoptosis.
