基于网络药理学探讨程序性坏死的中医病机及用药规律

Study on the pathogenesis and medication rules of procedural necrosis in Chinese medicine based on network pharmacology

摘要 目的探讨中药调控程序性坏死(Necroptosis,Nec)的病机特征、物质基础及用药规律。方法检索Gene Cards、OMIM、Drug Bank、KEGG、CTD、TTD获取Nec靶点,基于ADME在TCMSP中筛选候选化合物及中药,并构建相互作用网络,匹配各中药性味归经以行规律分析。结果共获652个Nec靶点,49个可匹配候选化合物的靶点,553个候选化合物和346味中药。干预Nec的药味为苦、甘、辛,药性以寒、温居多,归肝、肺、胃、脾、肾经。结论总结中药干预Nec相关成分及规律,推测Nec病机特征可能与气滞痰结、血瘀毒聚有关,治当行气开结、活血解毒。文中较系统地探讨中药干预Nec的物质基础和用药规律,为今后中西医结合临床治疗及干预Nec中药新药研发提供思路和依据。 Objective To investigate the pathogenesis characteristics,material basis and medication regularity of necroptosis(Nec)regulated by Chinese medicien.Methods Gene Cards,OMIM,Drug Bank,KEGG,CTD and TTD were searched to obtain Nec targets.Based on ADME,compounds and herbs acting on targets were selected in TCMSP,and the interaction networks were constructed to match the taste of each herbs for regular analysis.Results As a result,a total of652 Nec targets were obtained,49 targets that matched the candidate compounds,553 candidate compounds and 346 Chinese medicines.The medicinal taste of Intervention Nec is bitter,sweet and pungent,and the medicinal properties are mostly cold and warm,which belong to the liver,lung,stomach,spleen,and kidney meridians.Conclusion Summarize the relevant components and laws of traditional Chinese medicine intervention Nec,and speculate that the pathogenesis of Nec may be related to the stagnation of Qi,phlegm,blood stasis and toxin accumulation.The article systematically discusses the material basis and medication rules of traditional Chinese medicine intervention in Nec,and provides ideas and basis for the future clinical treatment of integrated traditional Chinese and western medicine and intervention in the development of new Chinese medicines for Nec.
