摘要 目的探讨miR-129-5p在卵巢癌中的表达及生物学作用。方法收集川北医学院附属医院手术切除的正常卵巢和癌组织。检测组织中miR-129-5p和TMEFF1的表达水平,并分析其相关性。将SKOV3细胞分为NC mimics组、miR-129-5p mimics组、NC inhibitors组、miR-129-5p inhibitors组和miR-129-5p mimics+TMEFF1组。检测miR-129-5p和TMEFF1 mRNA及蛋白的表达,进一步检测细胞的增殖、迁移和侵袭能力,并验证其靶向关系。结果卵巢癌组织中miR-129-5p的表达下调,TMEFF1的表达上调(P <0.01),两者呈负相关(r=-0.848,P <0.01),且均与淋巴转移和FIGO分期有关联(P <0.05)。过表达miR-129-5p降低SKOV3中TMEFF1 mRNA和蛋白的表达水平、抑制SKOV3增殖、迁移和侵袭能力(均P <0.01),而抑制miR-129-5p表达则呈现出相反的效果。过表达TMEFF1可逆转miR-129-5p mimics对SKOV3的抑制作用。结论 miR-129-5p调控TMEFF1抑制卵巢癌的进展。 Objective To investigate the expression level of miR-129-5p gene and its biological effects in ovarian cancer.Methods The normal ovaries and cancer tissues were collected from the Affiliated Hospital of North Sichuan Medical College.The expression levels of miR-129-5p and TMEFF1 were detected and their correlation was analyzed.The SKOV3 cells were divided into NC mimics group, miR-129-5p mimics group, NC inhibitors group, miR-129-5p inhibitors group and miR-129-5p mimics+TMEFF1 group.The mRNA and protein expressions of miR-129-5p and TMEFF1 were detected.The cell proliferation, migration and invasion ability were further detected, and the targeting relationship was verified.Results The expression of miR-129-5p in ovarian cancer tissue was down-regulated(P < 0.01), and the expression of TMEFF1 was upregulated(P < 0.01), and the expression of miR-129-5p and TMEFF1 was negative correlation(r=-0.848, P < 0.01).Both miR-129-5p and TMEFF1 are associated with the presence or absence of lymphatic metastasis and FIGO stage in patients with ovarian cancer(P < 0.05).Overexpression of miR-129-5p reduced the expression levels of TMEFF1 mRNA and protein in SKOV3, inhibited the proliferation, migration and invasion ability of SKOV3(all P < 0.01), while the inhibition of miR-129-5p expression showed the opposite effect.Overexpression of TMEFF1 reversed the inhibitory effect of miR-129-5p mimics on ovarian cancer SKOV3 cells.Conclusion miR-129-5p regulates TMEFF1 to inhibit the progression of ovarian cancer.
