摘要 对韩信草总黄酮的提取工艺进行优化,并研究其对4种供试菌的抑菌活性。通过超声波辅助醇提取法提取韩信草中的总黄酮,在单因素试验基础上,以液料比、超声时间、超声功率、乙醇浓度为影响因素,以韩信草总黄酮含量为响应值,应用Box-Behnken响应面法优选韩信草总黄酮的提取工艺条件。同时,采用滤纸片法测定韩信草总黄酮对4种供试菌抑菌活性。研究结果表明,韩信草总黄酮超声提取最佳工艺为:液料比为26mg/L、超声功率为307W、超声时间为20min、乙醇浓度为60%。在此条件下,韩信草总黄酮的含量为7.75mg/g。抑菌研究结果表明,经优化的最佳工艺下得到的韩信草总黄酮对金黄色葡萄球菌、铜绿假单胞菌、大肠埃希菌和白色念珠菌具有不同程度的抑菌作用。 In this paper,the extraction process of total flavonoids from Scutellaria indica was optimized,and its antibacterial activity against four tested bacteria was studied.Ultrasonic-assisted alcohol extraction method was used to extract total flavonoids from Scutellaria indica.On the basis of single factor experiment,the Box-Behnken response surface method was used to optimize the extraction conditions of total flavonoids from Scutellaria indica with liquid-solid ratio,ultrasonic time,ultrasonic power and ethanol concentration as the influencing factors,and the total flavonoids content of Scutellaria indica as the response value.At the same time,filter paper method was used to determine the antibacterial activity of the total flavonoids of Scutellaria indica against four kinds of tested bacteria.The results showed that the optimal extraction conditions were as follows:liquid/solid ratio was 26mg/L,ultrasonic power was 307W,ultrasonic time was 20min and ethanol concentration was 60%.Under these conditions,the content of total flavonoids was 7.75mg/g.The results of bacteriostatic study showed that the total flavonoids obtained by the optimized process had different degree of bacteriostatic effects on Staphylococcus aureus,Pseudomonas aeruginosa,Escherichia coli and Candida albicans.
