龙虎人丹对小鼠酒精性肝损伤的保护作用研究 被引量:3

Protection of LongHu RenDan on Alcoholic-Induced Hepatic Injury in Mice

摘要 目的:探究龙虎人丹对小鼠酒精性肝损伤的保护作用。方法:将36只C57小鼠随机分为6组:正常对照组、模型对照组、龙虎人丹20、40、80 mg/kg组及水飞蓟宾70 mg/kg组,每组6只。使用Lieber-DeCarli液体饲料喂养11 d后再给予单次乙醇灌胃来建立小鼠急性酒精性肝损伤模型。每日各组小鼠按相应剂量药物进行干预,第11 d正常对照组给予麦芽糖糊精,其余各组给予酒精灌胃,9 h后取血并收集肝组织,HE染色观察小鼠肝脏病理情况,检测小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)的活性和总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)以及肝组织中三酰甘油(TG)的含量,Western Blot检测肝组织中CYP2E1蛋白的表达水平。结果:与正常对照组相比,模型对照组小鼠血清中的ALT、AST活性、TC、 LDL含量均显著增高(P<0.01),HDL含量显著降低(P<0.01),肝组织出现明显的空泡,脂肪变性和细胞坏死。与模型对照组相比,龙虎人丹20、40、80 mg/kg能有效改善小鼠血清中的ALT、AST活性、HDL、LDL含量(P<0.05或P<0.01)。组织病理染色结果显示龙虎人丹能够改善肝脏内脂质堆积和炎症因子蓄积,肝脏内TG含量显著下降(P<0.01)。Western Blot结果显示,龙虎人丹可显著下调肝组织中酒精代谢关键酶细胞色素P450 2E1(CYP2E1)蛋白表达(P<0.01)。结论:龙虎人丹能够保护酒精诱导的小鼠急性肝损伤,具有解酒保肝的功效。 Objective: To investigate the hepatoprotective effects of LongHu RenDan on alcoholic-induced liver injury in mice. Methods: 36 C57 mice were randomly divided into 6 groups: the control group, the model group, LongHu RenDan groups(20 mg/kg、40 mg/kg and 80 mg/kg) and the positive control(silybin) group(70 mg/kg), with 6 mice in each group. Lieber-DeCarli ethanol diet was given for 11 days followed by gavage of a single dose of ethanol to establish the acute alcoholic-induced liver injury model. Mice were intervened by corresponding drugs once day, on 11 th-day, the mice in the control group were given maltodextrin and those in the other groups were given alcohol by gavage. 9 h after administration, serum and liver tissues were collected, liver tissue samples were stained by HE to examine the histopathological changes. Serum levels of alanine aminotransferase(ALT), glutamic-oxalacetic transaminease(AST), total cholesterol(TC), high-density lipoprotein(HDL),low-density lipoprotein(LDL) and hepatic tissue triglyceride(TG) level were detected. The expressions of Cytochrome P450 2 E1(CYP2 E1) in liver tissues were detected by Western blot. Results: Compared with the control group, the serum contents of ALT, AST, LDL and TC were significantly increased in the model group(P<0.01), HDL content was significantly reduced(P<0.01). In liver tissues, obvious vacuoles, steatosis and cell necrosis were observed. Compared with the model group, the levels of ALT, AST, HDL and LDL were improved in LongHu RenDan groups(20、40、80 mg/kg)(P<0.05 or P<0.01). Histopathological staining results showed that LongHu RenDan significantly reduced the lipid accumulation and the aggregation of inflammation factors. The content of TG in the liver was significantly decreased(P<0.01). Western blot results showed that the expression level of cytochrome P450 2 E1(CYP2 E1) in the alcohol metabolism was significantly down-regulated(P<0.01). Conclusion: LongHu RenDan can protect mice from acute alcohol-induced liver injury. It has Jiejiubaogan effect.
